Objective: The NASH model was successfully established by feeding SD rats with a high-fat diet to investigate the levels of serum endotoxin and IL-28 B in nonalcoholic steatohepatitis.Methods: Thirty-eight male SD rats were selected and divided into 2 groups according to body weight,namely,the control group(12 rats),and given normal dietary feed;the intervention group(26 rats),were given high-fat dietary feed.SD rats fed with high-fat diet can reach moderate to severe steatohepatitis in 8-12 weeks of liver pathology.Our preliminary experiment found that the liver pathological changes of SD rats reached NASH at the end of the 12 th week.In this experiment,at the 12 th week,2rats in the intervention group were randomly taken out and sacrificed for liver histopathological examination,which also proved that the liver pathological changes reached the NASH diagnosis.,Indicating that the NASH modeling of SD rats was successful.Twelve SD rats were randomly selected in the intervention group and sacrificed together with the control group of 12 rats.After fasting for 12 hours before sacrifice,the SD rats were weighed.The whole blood of each rat was collected and serum was obtained by centrifugation to detect ALT.,AST,GGT,TG,TC,LDL,ET,IL-28 B levels,dissect the abdominal cavity and remove the liver,measure the liver wet weight and calculate the liver index,and retain the liver specimens for pathological examination.At the beginning of 13 weeks,the remaining 12 SD rats in the intervention group changed the high-fat dietary feed to the ordinary dietary feed.Four rats died at the end of the 13 th week,14 th week,and 15 th weekend respectively,and were weighed 12 hours after fasting.The weight of SD rats,the whole blood of each rat was collected and the serum was obtained by centrifugation,the levels of ALT,AST,GGT,TG,TC,LDL,ET,IL-28 B were detected,the liver index was calculated and the liver histopathological examination was performed.SPSS 22.0 statistical analysis software was used for data analysis.The body weight,liver index,liver pathological score,and serum ALT,AST,GGT,TG,TC,LDL,ET,IL-28 B level changes and analysis of the correlation between liver pathological score and ET,IL-28 B.After the intervention group began to switch to regular diet for 13 weeks,4 animals were sacrificed each week to compare body weight,liver index,liver pathological score,and serum ALT,AST,GGT,TG,TC,LDL,ET,IL-28 B levels.P <0.05 was considered statistically significant.Results:(1)The liver pathological results of the intervention group at the 12 th weekend showed: diffuse and moderate steatosis of the liver lobule,partial nucleus disappearance and necrosis and dissolution of some hepatocytes,hepatic sinus space disappeared,a large number of inflammatory cells infiltrated in the manifold area,and fibrosis was obvious.The pathological results of the liver in the control group showed that the liver lobule structure was complete,the hepatocytes were arranged neatly along the central vein,the cell morphology was regular,the boundaries between the cells were clear,the hepatocyte nucleus was centered,the hepatic sinus space was clear,and there was no inflammation infiltration and fibrosis formation.Compared with the control group,the intervention group had normal liver tissue destruction,obvious liver fibrosis,hepatocyte necrosis,liver inflammatory cell infiltration,and significant lipid accumulation in the liver.(2)The body weight of the intervention group was lower than that of the control group,the liver index was higher than that of the control group,and the liver pathological change score was higher than that of the control group,the difference was statistically significant(P <0.05).(3)The levels of serum ALT,AST,GGT,TG,TC,LDL,ET in the intervention group were higher than those in the control group,while the levels of IL-28 B were lower than those in the control group,and liver pathology was positively correlated with endotoxin levels(r = 0.86),Negatively correlated with IL-28 B level(r =-0.67),the difference was statistically significant(P<0.05).(4)After the intervention group changed the high-fat feed to ordinary feed,the liver pathological results at the end of the 15 th week showed that the liver lobule was diffuse and mild steatosis,and hepatic cells were almost completely regenerated,and hepatic cord,hepatic plate and hepatic sinusoidal space appeared.The liver plate arrangement was slightly disordered,no infiltration of inflammatory cells was seen in themanifold area,and fibrosis disappeared.The pathological results on the 13 th weekend showed: diffuse and moderate steatosis of hepatic lobule,hepatocyte regeneration,hepatic plate and hepatic sinusoidal spaces were seen,and only a small amount of inflammatory cell infiltration was seen in the junction area,interfacial inflammation,and obvious fibrosis.At 15 weeks,liver fibrosis disappeared,liver cells regenerated,a small amount of inflammatory cells infiltrated in the liver,and a small amount of fat accumulated.(5)Compared with 13,14 and 15 weeks at 12 weeks,body weight increased compared with before,liver index decreased compared with before,and liver pathological score decreased compared with before,the difference was statistically significant(p <0.05).At 13 weeks compared with 14,15 weeks,the serum ALT level was significantly lower than before;at 13 weeks compared to 14,15 weeks,the serum AST level was significantly lower than before;compared between 13,14,15 weeks,the serum GGT level Significantly lower than before,the difference was statistically significant(p <0.05).Compared with 14 and 15 weeks,serum TG levels were significantly lower than before;13 weeks and 15 weeks,serum TC levels were significantly lower than before;13,14 and 15 weeks,serum LDL levels were significantly higher than before.Elevated,the difference was statistically significant(p<0.05).At 13 weeks,compared with 14,15 weeks,serum endotoxin levels were significantly lower than before;at 13 weeks,compared with 14,15 weeks,serum IL-28 B levels were significantly higher than before,the difference was statistically significant(p<0.05).Conclusion:1.Non-alcoholic steatohepatitis SD rats induced by high-fat diet can lead to increased serum endotoxin levels and decreased serum IL-28 B levels.2.Pathological changes of non-alcoholic fatty liver disease caused by high-fat diet are positively correlated with serum endotoxin levels and negatively correlated with serum IL-28 B levels.It is speculated that IL-28 B may be a protective factor for non-alcoholic fatty liver disease.
