Objective To verify the adsorbability of nano-hydroxyapatite(nHA)surface,test its adsorption ability for anti-P.gingivalis-IgY,and the slow-release process of antiP.gingivalis-IgY in vitro,so as to build nano-hydroxyapatite composite material with anti-P.gingivalis-IgY slow-release function,and finally test the composite material in vitro with the ability of P.gingivalis,the composite material with antibacterial,sustained-release and biocompatibility was successfully constructed,which provides theoretical basis for the future clinical application.Methods The structure of nano-hydroxyapatite was observed by SEM.The yolk antibody against Porphyromonas gingivalis was loaded on nHA,and the ability of nano-hydroxyapatite loaded with IgY protein was verified by FT-IR,and the drug release behavior of IgY-nHA composite was also tested.Porphyromonas gingivalis was selected as the experimental strain to study the minimum inhibitory concentration of IgY-nHA composite and the inhibitory effect of different concentrations of IgY-nHA composite on the growth of P.gingivalis.Results 1.SEM examination shows that nHA is a porous structure composed of nano-particles;2.The results of FT-IR showed that nHA successfully loaded IgY protein;3.With the increase of the concentration of IgY protein loading solution,the adsorption capacity of nHA to IgY protein also increased significantly.When the concentration of IgY protein solution is 4mg/ml,the adsorption capacity of nHA to IgY protein is653mg/g;4.In the first one hour,the release of IgY protein from nHA solution was 16%of the total amount of adsorption.At this stage,the main release was the weak binding IgY protein on the surface of nHA.In the next 36 hours,IgY protein was released slowly and continuously,and then reached the release platform stage.The percentage of IgY protein released at 96 h was 34%;5.The minimum inhibitory concentration of IgY-nHA to P.gingivalis was 6mg/ml;6.The curve of the effect of IgY-nHA on the growth of P.gingivalis shows that IgY-nHA can delay the time point of P.gingivalis entering the logarithmic growth period and inhibit the growth of P.gingivalis in a concentration dependent manner.Conclusion nHA has a good ability of adsorption and release of IgY protein,and IgY nHA can inhibit the proliferation of P.gingivalis,which has a great application prospect in the treatment and prevention of periodontal disease.
