Schizophrenia(SZ)is known to be a mental illness with unknown causes and different symptoms, and genetic factors play an important role in the pathogenesis of schizophrenia.Studies have shown that there are abnormal connections between the Salience Network(SN),the Central Executive Network(CEN),and the Default Mode Network(DMN)in many mental and neurological diseases including schizophrenia. We collectively refer to these three sub-networks as the brain’s core neural cognitive network, which the salience network plays a role in regulating the switching between the central execution network and the default network. But whether the abnormality of the brain’s core cognitive network in patients with schizophrenia is caused by congenital or acquired, there is no definite conclusion so far.The purpose of this paper is to study the problem in depth by using methods such as Independent Component Analysis(ICA)and the construction of a seed point whole brain functional connection network. The study was conducted in 20 schizophrenia patients(SZ),23 siblings of schizophrenia patients(SB)and 33 healthy controls(HC).We use spatial independent component analysis to construct the core cognitive network of the brain, and further obtain the time series of the three sub-networks.Based on the regulatory role of the salience networkin the core neural cognitive network,we define the difference between the correlation value of the SN and CEN time series and the correlation value of the SN and DMN time series as the Network Interaction Index(NII)and calculate Static NII value, dynamic NII value, and NII value in different states and make an inter group comparison.Next,we further use the above-mentioned brain core neural cognitive network as a seed point to establish a functional connection network with other voxels in the whole brain.Statistical analysis is performed through methods such as analysis of variance and two-sample T test,to find out the significant differences between the groups of clusters which the correction has passed,so as to further explore the pathological mechanism of SZ.The conclusions of this paper are two: First,we found that the NII values of patients was significantly lower than healthy controls, whether static or dynamic, but the NII values of patients and siblings, and NII values of siblings and healthy controls were not found to have significant differences.Second,we found that there were significant differences in brain regions among the three groups of subjects. The connections in patients with brain regions such as the temporal lobe were different from those in normal subjects; The connections in patients with brain regions such as the lentiform nucleus were different from those in their siblings; The connections in siblings with brain regions such as the caudatenucleus were different from those in normal subjects. This paper combines the internal connections of the brain’s core neural cognitive network and the whole brain functional connection to study the disease mechanism and genetic mechanisms of schizophrenia. The obtained results show that although the network abnormalities of SZ patients are genetically related, they are also affected by external environmental factors. It has certain reference and guidance significance for the clinical treatment and diagnosis and prevention of schizophrenia.
