Objective:To observe the effects of combined acupuncture and rehabilitation on neuroethology in rats with hypoxic-ischemic brain damage,and to study the effects of combined acupuncture and rehabilitation on apoptosis and expression of apoptosis-related proteins AIF and Fas in cerebral cortex of HIBD rats The study provided a basis for the treatment of HIBD through apoptotic pathway by acupuncture combined with rehabilitation.Methods:A total of 160 7-day-old Wistar rats were selected,they were randomly divided into a sham operation group,a model group,an acupuncture group,a rehabilitation group,and an acupuncture combined with rehabilitation group.Each group was further divided into 4 subgroups at 12 h,24h,48 h,and 72 h,with 8 in each group.The modified Rice method was used to establish the HIBD model.The rats in the sham operation group and the model group were not treated.The acupuncture group was treated with scalp acupuncture.The rehabilitation group was subjected to cage and rod training.The acupuncture combined rehabilitation group performed cage and rod training at the same time as the scalp acupuncture.Rats in each group were evaluated with modified Bederson scores at various time points after treatment.After decapitation,TUNEL was used to detect apoptosis in rat cerebral cortex,and RT-PCR was used to detect rat cerebral cortex.AIF and Fas gene expression.Results:1.Neurological deficit score: Compared with the sham operation group,the neurological deficit scores of rats in each group at each time point have significant differences(P<0.05);compared with the model group,each treatment group has The neurological defect scores at each time point were significantly reduced,and the differences were significant(P<0.05).Compared with the acupuncture group and the rehabilitation group,the acupuncture combined with rehabilitation group had no significant differencein neurological deficit score at 12h(P>0.05).At the three time points of 24 h,48h,and 72 h,acupuncture combined with rehabilitation group’s neurological deficit score compared with the neurological deficit scores of the acupuncture group and the rehabilitation group,the difference was significant(P<0.05).2.Apoptosis in the cerebral cortex: Compared with the sham operation group,the number of positive cell apoptosis at each time point in each group of rats has a significant difference(P<0.05);compared with the model group,each The number of positive apoptotic cells in the treatment group was significantly reduced at each time point,and the difference was significant(P<0.05).At each time point,the number of positive cell apoptosis in the acupuncture combined with rehabilitation group was also significantly different from that in the acupuncture group and the rehabilitation group(P<0.05);except for the sham operation group,the number of apoptosis at each time point in each group showed a peak change,that is,apoptosis at 12 h The number gradually increased,the number of apoptosis reached its peak at24 hours,and the number of apoptosis gradually decreased at 48 and 72 hours.3.AIF and Fas m RNA expression in the cerebral cortex: Compared with the sham operation group,the expression of AIF and Fas m RNA at each time point in each group of rats has significant differences(P<0.05);compared with the model group In comparison,the expression levels of AIF and Fas m RNA in each treatment group at each time point were significantly reduced,and the differences were significant(P<0.05);At two time points,24 h and48h,the AIF and Fas m RNA expression levels in the acupuncture combined with rehabilitation group were significantly different from those in the acupuncture group and rehabilitation group(P<0.05);Except for the sham operation group,the expression levels of AIF and Fas m RNA at each time point in each group also showed a peak-like change,and the trend was the same as that of apoptosis.Conclusion:1.Acupuncture combined with rehabilitation method can improve theneurological deficit score of HIBD rats,and is superior to single acupuncture method or rehabilitation method;2.Acupuncture combined with rehabilitation method can regulate the neuronal apoptosis in the cerebral cortex of HIBD rats,and is superior to single acupuncture method or rehabilitation method;3.Acupuncture combined with rehabilitation method may reduce the expression levels of AIF and Fas m RNA in the cerebral cortex of HIBD rats and inhibit cell apoptosis.
