Objectives To explore the therapeutic effect and mechanism of Leonurus chinensis combined with Astragalus membranaceus on type Ⅲprostatitis in rats,and to provide theoretical basis and experimental basis for its further experimental research and clinical application in the future.Methods The rats were randomly divided into control group,model group,Qianliakang group,motherwort group,astragalus membranaceus group and combined group.Except for the control group,the rats in the other groups were established by prostate injection of carrageenan.Leonurus officinalis group and Astragalus membranaceus group were given2g/(kg·d)and 2g/(kg·d)of Leonurus officinalis and Astragalus membranaceus 2g/(kg·d),respectively.The combined dose(1g/(kg·d),1g/(kg·d))of Astragalus membranaceus)was given intragastrically.The dose of 0.76g/kg was given intragastrically(6.3 times as much as the commonly used clinical dose),and the blank group was given saline intragastrically.Before and 28 days after administration,the body weight and urination volume of the rats were measured,and the mechanical pain threshold of the rats was measured after the last intragastric administration.The(EPS)of the rat prostate massage fluid was taken on the slide,the color and transparency of the liquid were observed with the naked eye,and the number of white blood cell(WBC)and the density of lecithin bodies in EPS were observed under microscope.The prostate,spleen and thymic gland were weighed and the organ index was calculated.The activity of SOD(SOD)and the level of malondialdehyde(MDA)in prostate homogenate and serum were measured by pathological section.The contents of tumor necrosis factor-a(INF-a)and IL-10(IL-10)in prostate homogenate and serum were detected by enzyme-linked immunosorbent assay(Elisa).The level of Zn2+ was measured by BH5N1 five-channel atomic absorption spectrometer.Results Compared with the control group,the rat model of type ⅢA prostatitis induced by carrageenan injection was successful.compared with the control group,the body weight and urination volume of the model group were significantly decreased,the pain threshold was significantly decreased,the number of leukocyte(WBC)in EPS was significantly increased,the score of lecithin corpuscle was significantly decreased,the score of prostate tissue lesion was significantly increased,and the coefficient of prostate and spleen was significantly increased.The(SOD)value of SOD in prostate tissue and serum was significantly decreased,the content of malondialdehyde(MDA)(MDA)was significantly increased,the level of tumor necrosis factor-a(TNF-α)was significantly increased,the level of IL-10(IL-10)was significantly decreased,and the level of Zn2+in prostate tissue was significantly decreased(P<0.01).Compared with the model group,the body weight of the four administration groups was significantly higher than that of the model group(P<0.05 or P<0.01),and the urination volume of the four administration groups was significantly higher than that of the model group(P<0.01),and the threshold of the four administration groups was significantly higher than that of the control group(P<0.05 or P<0.01).The prostate coefficient of Qianliankang group,motherwort group and combined group was significantly lower than that of Astragalus membranaceus group(P<0.01),but there was no significant difference between astragalus membranaceus group and Astragalus membranaceus group(P>0.05).The spleen coefficient of Qianliankang group and combined group was significantly lower than that of control group(P<0.05 or P<0.01),and the spleen coefficient of Leonurus chinensis group and Astragalus membranaceus group had a decreasing trend,but the difference was not statistically significant(P>0.05),and the number of leukocyte(WBC)in EPS of the four administration groups was significantly decreased(P<0.05 or P<0.01),and there was no significant difference in spleen coefficient between the four treatment groups(P<0.05 or P<0.01).The scores of lecithin corpuscles in EPS of Qianliankang group,motherwort group and combined group were significantly higher than those of astragalus membranaceus group(P>0.05),and the scores of prostate pathological changes in the four treatment groups were significantly lower than those in the control group(P<0.05 or P<0.001),and the scores of lecithin corpuscles in Astragalus membranaceus group were significantly higher than those in the control group(P<0.05 or P<0.05),and the scores of lecithin corpuscles in Astragalus membranaceus group were significantly higher than those in the control group(P<0.05).In prostate tissue and serum,the(SOD)values of SOD in the four groups were significantly higher than those in the control group(P<0.005 or P<0.01),and the SOD levels in the four groups were significantly higher than those in the control group(P<0.05 or P<0.01).In prostate tissue and serum,the content of malondialdehyde(MDA)(MDA)in prostate tissue and serum of Qianliankang group,motherwort group,combined group was significantly lower than that of control group(P<0.05 or P<0.01).The content of(MDA)in astragalus membranaceus group had a downward trend,but there was no significant difference(P>0.05).In prostate tissue and serum,the levels of IL-10(IL-10)in Qianliankang group,motherwort group and combined group were significantly higher than those in Astragalus membranaceus group(P<0.05 or P<0.01),but there was no significant difference in IL-10 level in Astragalus membranaceus group(P>0.05).In prostate tissue and serum,the level of tumor necrosis factor-a(TNF-α)in Qianliankang group,motherwort group and combined group was significantly lower than that in astragalus group(P<0.05 or P<0.01),but there was no significant difference in tumor necrosis factora(TNF-α)in astragalus membranaceus group(P>0.05).In prostate tissue,the level of Zn2+in Qianliankang group and combined group was significantly higher than that in control group(P<0.05 or P<0.01).Although the level of Zn2+ in Leonurus group and Astragalus membranaceus group was increased,there was no significant difference(P>0.05).Among the treatment groups,the changes of the indexes in the combined group were the most obvious,and there were significant differences in other indexes except the content of malondialdehyde(MDA)and body weight(P<0.05 or P<0.01).Conclusions The combination of Leonurus chinensis and Astragalus membranaceus has obvious therapeutic effect on rat model of type IIIA prostatitis,among which the combined group is the most effective.It is speculated that the mechanism may be related to diuresis,anti-inflammation,antioxidation,regulation of immunity and increase of
