Objective:To study the changes of lncrna in enterocytes of rats infected with Escherichia coli.To find out the lncrna related to cell infection and verify its function.Methods:The best concentration and time of E.The expression of NONRATT006155.2 was detected by quantitative PCR,and the expression of MAP4K3 and β-defensin-2 at protein level was detected by Western blot.MTT was used to detect the inhibition of the cells.Results:The inhibition rate of 103 E.coli / ml was the best at 24 hours.By sequencing analysis,it was found that NONRATT006155.2 decreased significantly,which was related to the expression of MAP4K3 and might affect the expression of defensin.The IEC-6overexpression cells of NONRATT006155.2 were successfully constructed.Quantitative PCR showed that the expression of NONRATT006155.2 was the highest in IEC-6 cells transfected with NONRATT006155.2 mimic plasmid,which was significantly higher than blank control group(without any treatment),E.coli group(only treated by E.coli)and E.coli + NC group(E.coli sensitive The cells which had been transfected with empty plasmids were stained,P<0.05).In E.coli group and E.coli+NC group,the expression of NONRATT006155.2was significantly lower than that in the blank control group(P<0.05).In the large intestine+mimic group,the inhibition rate of cells gradually decreased with the increase of action time(P<0.05),which was significantly lower than that of the large intestine group and the large intestine + NC group at the same time point(P<0.05).By western,the expression of MAP4K3 and β-defensin-2 in the protein level was the lowest in the large intestine + mimic group,significantly lower than the other three groups(P<0.05),and the highest in the large intestine group and the large intestine+NC group,significantly higher than the blank control group(P< 0.05),but there was no significant difference between the large intestine group and the large intestine+NC group(P>0.05).Conclusion:The expression of NONRATT006155.2was negatively correlated with the expression of MAP4K3 and β-defensin-2 in IEC-6 cells.Therefore,NONRATT006155.2 may be involved in the regulation of MAP4K3 and β-defensin-2 expression.
