Objective By establishing the model of type 2 diabetes mellitus(T2DM)rats,we observed the effect of Dahuang Huanglian Xiexin Decoction on the changes of blood glucose,blood lipid,pancreatic histopathology of T2DM rats and the m RNA and protein expression of AMP-acticated protein kinase α(AMPK α),Peroxisome proliferator activated receptor-γ gamma coactivator-1α(PGC-1 α)and glucose transporter-4(GLUT4)in skeletal muscle of T2DM rats,To explore the hypoglycemic and lipid-lowering effects of Dahuang Huanglian Xiexin Decoction on T2DM rats,and to explore its mechanism,so as to provide experimental basis for the treatment of T2DM with traditional Chinese medicine.Methods According to the random number table method,40 healthy SPF male Wister rats were divided into the blank control group with 10 rats and the model group with 30 rats.The blank control group was fed with normal diet,and the model group was used to prepare the T2DM rat model by high-sugar and high-fat feed combined with low-dose streptozotocin(STZ)method.After successful modeling,the rats were divided into model group,traditional Chinese medicine group and metformin group according to the random number table method,with 10 rats in each group.The traditional Chinese medicine group was given Dahuang Huanglian Xiexin Decoction with a dose of 1g/(100 g·d).The metformin group was given metformin by gavage with a dose of 18 mg/(100 g·d).The blank control group and model group were given the same amount of saline,and the drug intervention was conducted for 4 weeks.After the experiment,blood samples were collected from the abdominal aorta,and serum samples were collected to determine fasting blood glucose(FPG),total cholesterol(TC),triglyceride(TG)and low-density lipoprotein cholesterol(LDL-C).Pancreatic tissues were taken from each group and pathomorphological changes were observed under optical microscope.RT-PCR was used to detect m RNA expressions of AMPKα,PGC-1α,GLUT4 in skeletal muscle of rats.The expressions of AMPKα,phosphorylated AMP-acticated protein kinase α(P-AMPKα),PGC-1α and GLUT4 were detected by Western Blot.Result After modeling,compared with the blank control group,FPG in the model group was significantly increased(P<0.01),which means models were induced successfully.With gavaging Dahuang Huanglian Xiexin Decoction for 4 weeks,compared with the model group,the traditional Chinese medicine group of FPG,TC,TG,LDL-C significantly decreased(P < 0.01 or P < 0.05);Under optical microscope,islet structure was still complete,the degeneration of islet cell β was light and the number of physaliphore reduced;the m RNA and protein expressions of AMPKα、PGC-1α、GLUT4 obviously increased in skeletal muscle(P<0.01 or P<0.05).Conclusion 1.Dahuang Huanglian Xiexin Decoction can reduce blood glucose and lipid in T2DM rats,regulate glucose and lipid metabolism,maintain glucose and lipid homeostasis in rats,and improve damaged islet cells.2.Dahuang Huanglian Xiexin Decoction can regulate the AMPK signaling pathway,and by up-regulating the expression of PGC-1α and GLUT4,it can improve insulin resistance(IR)in skeletal muscle of T2DM rats.This is one of its mechanisms for treating T2DM.
