Objectives This study aims to investigate the relationship between single nucleotide polymorphism in the promoter of tumor necrosis factor superfamily 15(TNFSF15)and the susceptibility to colorectal cancer.Methods The study used the group-based case-control method.The case group involved in 700 cases of colorectal cancer and 700 healthy controls collected.Using restriction fragment length polymorphism polymerase chain reaction(PCR-RFLP)technology for genotyping.Two SNPs(rs6478109,rs7848647)of TNFSF15 were selected as targets.We used ?2 test to compare the distribution differences of the two group between gender,age,smoking and drinking status,and then analyze the effect of different genotypes on the degree of cell differentiation.OR and 95% confidence interval(CI)we calculated by unconditional logistic regression to evaluate the relationship between these two SNPs and the risk of colorectal cancer.Results Our data showed that the individuals with TNFSF15 rs6478109 TC or CC genotype have increased risk of developing colorectal cancer compared with the TT genotype carriers(OR=1.52,95%CI 1.20-1.91;OR=1.75,95%CI 1.30-2.37).For TNFSF15 rs7848647 polymorphism,GG genotype was associated with the increased risk of colorectal cancer compared with AA genotype(OR=1.76,95%CI 1.30-2.39).We then conducted stratified analysis by gender,age,smoking and drinking status.We found that TNFSF5 rs6478109 CC genotype decreased colorectal cancer risk among male(OR=0.51,95%CI 0.35-0.75),eldly(age>60)(OR=0.47,95%CI 0.31-0.72),non-smoker(OR=0.46,95%CI 0.33-0.66),and non-drinker(OR=0.57,95%CI 0.41-0.80).We also demonstrated that rs7848647 GG genotype reduced the genetic susceptibility to colorectal cancer among males(OR=0.53,95%CI 0.36-0.77),youngers(age?60)(OR=0.50,95%CI 0.32-0.77),eldly(age>60)(OR=0.62,95%CI 0.40-0.95),non-smoker(OR=0.57,95%CI 0.40-0.81),drinker(OR=0.45,95%CI 0.23-0.88),and non-drinker(OR=0.58,95%CI 0.41-0.82).No evidence of correlation between two single nucleotide polymorphisms and colorectal cancer cell differentiation.Conclusions TNFSF15-358T>C(rs6478109)and-638A>G(rs7848647)polymorphisms affects the genetic susceptibility to colorectal cancer.Figure 2;Table 10;Reference 192...
