Objective: By observe the effect of external application of chick embryo egg on the expression of bone morphogenetic protein-2(BMP-2)and insulin-like growth factor-1(IGF-1),and to explore the mechanism of external application of chick embryo egg on the repair of bone defects in rats with nonunion of femur.Methods: 35 SD rats were selected,10 rats in the blank group,25 rats in the model group and 5 rats verify the model.The remaining model rats were randomly divided into treatment group(external application of chick embryo egg)and control group with 10 rats in each group.At 3,8 weeks after intervention,5 rats in the treatment group and control group were randomly selected for X-ray examination to observe the formation of callus,HE staining to observe the Histomorphological of bone defects under light microscope;The enzyme-linked immunosorbent assay(ELISA)was used to determine the changes of BMP-2 and IGF-1 in soft tissue of fracture site in blank group,control group and treatment group 3,8 weeks after intervention.Results: X-ray examination: after 3 weeks of intervention,the fracture line of the fracture end of the control group was clear,some periosteal reaction,no obvious callus and bone bridge were found.and a little callus shadow was found in the treatment group,and the fracture line was blurred than that of the control group.After 8 weeks of intervention,the fracture line of the broken end of the fracture in the control group was obvious,a small amount of callus formed,no bone bridge was formed and the marrow cavity was not open,while in the treatment group,the broken end of the fracture was formed,the marrow cavity was recanalized,and the plasticity of the fracture end was good(P < 0.01).Histomorphological examination: after 3 weeks of intervention,there was no obvious bone healing in the control group and the treatment group,but the repair trend of bone tissue in the treatment group was better than that in the model group.After 8 weeks of intervention,the areas of bone nonunion in all control group did not heal.In the treatment group,the bone tissue was basically intact in 4 cases,and only 1 case showed delayed healing.The content of BMP-2 and IGF-1: after 3 weeks of intervention,the concentrations of BMP-2 and IGF-1 in the experimental group were significantly higher than those in the control group and the blank group,and after 8 weeks,the BMP-2 in the blank group was higher than that in the control group,but there was no significant difference between the blank group and the experimental group(P > 0.05),and there was no significant difference in IGF-1 among the three groups after 8 weeks.Conclusion: external application of chick embryo egg can promote bone healing of SD model rats with nonunion of femur and regulate the concentration of BMP-2 and IGF-1 in soft tissue of bone defect area.
