| Objective To investigate the effect and mechanism of high mobility group protein B1-IL-1βcomplex on the expression of tumor necrosis factor-αand interleukin-6 in synovial fibroblasts of rheumatoid arthritis.Methods The synovial tissues of 10 patients with rheumatoid arthritis were isolated and cultured by digestive enzyme culture,and the fifth generation of rheumatoid synovial fibroblasts were used in the experiment.Stimulation of synovial fibroblasts from rheumatoid arthritis with different concentrations of HMGB1(0.1、1、10、100ng/m1)、IL-1β(0.01、0.1、1、10 ng/m1)、HMGBl/IL-1βcomplex(0.1/0.01、0.1/0.1、0.1/1、0.1/10、1/0.01、1/0.1、1/1、1/10、10/0.01、10/0.1、10/1、10/10、100/0.01、100/0.1、100/1、100/10 ng/m1).Detection of tumor Necrosis Factor-αand Interleukin-6 m RNA expression in synovial fibroblasts of Rheumatoid Arthritis by Real-time quantitative Polymerase chain reaction.Detection of TNF-αand IL-6 in the supernatant of synovial fibroblasts from patients with rheumatoid arthritis by enzyme-linked immunosorbent assay.Results Compared with the control group,the expression level of TNF-αm RNA and concentration of TNF-αin the IL-1βintervention group were higher than those in the control group(P<0.05);The expression of TNF-αm RNA and concentration of TNF-αin HMGB1/IL-1βcomplex intervention group was higher than that in HMGB1 intervention group(P<0.05);Compared with HMGB1 intervention group,the expression of TNF-αm RNA and concentration of TNF-αwas increased in HMGB1/IL-1βcomplex intervention group with different concentration gradients(P<0.05);Compared with the IL-1βintervention group,the expression of TNF-αm RNA and concentration of TNF-αincreased when IL1βincreased to 1ng/ml and10ng/ml in each concentration gradient HMGB1/IL-1βcomplex intervention group,and the expression of TNF-αm RNA increased in a dose dependent manner with the increase of HMGB1 concentration(P<0.05);Compared with the control group,the expression level of IL-6 m RNA and concentration of IL-6 in the IL-1βintervention group were higher than those in the control group(P<0.05);The expression of IL-6m RNA and concentration of IL-6 in HMGB1/IL-1βcomplex intervention group was higher than that in HMGB1 intervention group(P<0.05);Compared with HMGB1 intervention group,the expression of IL-6 m RNA and concentration of IL-6 was increased in HMGB1/IL-1βcomplex intervention group with different concentration gradients(P<0.05);Compared with IL-1βintervention group,the expression of IL-6 m RNA and concentration of IL-6 was increased in HMGB1/IL-1βcomplex intervention group with different concentration gradients Conclusion 1.The formation of HMGB1-IL-1βcomplex can promote the secretion of TNF-αand IL-6,by RASFs,but the inflammatory ability of HMGB1 itself is very weak,and the induction of RASFs is weaker than that of IL-1β,indicating that HMGB1,as a co-inflammatory factor,cooperates with IL-1βto mediate the expression of TNF-αand IL-6 by RASFs.2.When the concentration of IL-1βin HMGB1-IL-1βcomplex increased to a certain range,the expression of TNF-αand IL-6 in RASFs increased in a dose dependent manner with the concentration of HMGB1. |
