Objective Gallbladder cancer is one of the common tumors of the digestive tract.It is characterized by insidious onset,no specific symptoms,and it is not easy to detect in the early stages of the disease.This study investigated the changes in the methylation level of the cysteine dioxygenase-1(CDO1)gene promoter in cancer tissues and serum of patients with gallbladder cancer,and provided reference experimental data for the early diagnosis of gallbladder cancer.Method Under sterile conditions,tissues and serum samples from patients with gallbladder cancer collected and confirmed in the hospital were collected.After treatment of the cancer tissue,10 mg of the central tissue of the patient's tumor specimen was taken.Before surgery,the patient collected 4 ml of venous blood on an empty stomach in the morning and injected it into a 5 ml negative pressure blood collection tube containing a coagulant.Centrifuge at 3 000 rpm for 5 minutes.Use a disposable pipette to slowly aspirate the upper serum and seal it in a 2 ml EP tube.After labeling,store it in a liquid nitrogen storage tank immediately.Tissue samples were digested with tissue lysate and proteinase K,and DNA was obtained using Promega's DNA IQ ? system.Serum samples were extracted with Biog serum free DNA extraction kit.Finally,the Methyl CDO1 TM Bisulfite conversion Kit kit was used to modify the DNA with sodium bisulfite.After PCR amplification,CDO1 gene promoter methylation test results were obtained.Samples from patients with gallbladder polyps and healthy patients were processed in the same way.Results After statistically processing the experimental results,it was found that the univariate analysis resultsshowed that the degree of CDO1 methylation in the tissues and serum of patients with gallbladder cancer was related to the patient's age,gender,and tumor stage,with P <0.05,which was statistically significant;The tumor markers CEA,CA125,and CA19-9 were correlated,with P <0.05,which was statistically significant;there was no correlation with the histological type of the tumor,P> 0.05,without statistical significance;CDO1 in tissue samples from patients with gallbladder cancer The methylation rate was 63.3%(19/30),and the corresponding serum sample CDO1 methylation rate was 60%(18/30).There was no CDO1 methylation in the tissue samples of patients with gallbladder polyps,and the CDO1 methylation rate in plasma samples was 6.6%(2/30,the difference between the tumor and the control group was statistically significant(P <0.05).Conclusions Patients with gallbladder cancer have abnormal methylation status of CDO1 gene promoter region in their cancer tissues and their serum,and their specificity is high.The detection of abnormal methylation of the CDO1 gene promoter region in serum DNA in the early diagnosis,recurrence monitoring and prognosis of gallbladder cancer may provide a more comprehensive feasible solution and ideas for the treatment of gallbladder cancer.
