Objective TO establish a primary rat hippocampal neuron culture system in vitro,and to investigate the effect of Sema3 F on the expression of Specific Protein-1 in primary rat hippocampal neurons,so as to provide a theoretical basis for the exploration of Sema3 F primordities.Methods newborn Wistar rats were selected within 24 hours and the hippocampal tissue was extracted for primary culture.The cells cultured for 72 hours were divided into experimental group and control group.In the experimental group,Sema3 F with a final concentration of 10ng/ml was added for 0min,5min,15 min and 30 min respectively.Control group:the same concentration of fetal bovine serum was added.The Protein expression of Specific Protein-1 was detected by Western Blot.SPSS22.0 statistical software was used for statistical analysis,and P<0.05 was considered statistically significant.Results In the primary rat hippocampal neuron culture system,compared with the control group,there was no significant Change in the expression of Specific protein-1 protein expression in the experimental group at 4 different time points,and there was no significant change in the expression of Specific Protein-1 at different time points in the control group.Conclusion Sema3 F has no significant effect on the expression of Specific Protein-1 in primary rat hippocampal neurons.
