There are 7 species and 3 varieties of Lycium L.in the north and northwest of China,most of which have a long history as a source of Chinese herbal medicine.Lycium barbarum L.is widely used in traditional Chinese medicine,its dry root bark is called Lycii Cortex,and gouqizi(Lycii Fructus)refers to the dried mature fruit of Lycium barbarum L..Modern research shows that there are many natural active ingredients in Lycium barbarum L.,such as alkaloids,peptides,flavonoids and so on.In terms of pharmacological effects and clinical applications,it has been found that Lycium barbarum L.can be used for anti-tumor,anti-oxidation and anti-aging.This subject systematically studied the chemical constituents of dried root and stem of Lycium barbarum L..The content and results are as follows.By comprehensive use of TLC,silica gel,ODS,Sephadex LH-20 dextran gel chromatography and analysis,semi-preparative high-performance liquid phase and other modern chromatographic separation and purification methods,37 compounds were isolated from ethanol extract of the root and stem from Lycium barbarum L.,and their structures were determined by spectroscopic method including NMR,mass spectrometry,circular dichroism,infrared,ultraviolet,as well as other spectroscopy methods,combining with other physical,chemical data and computitional ECD method.The structures of these compounds were determined as (1R,4R)-hydroxy-2-azabicyclo[2.2.2]oct-5-en-one(1*),N-trans-feruloyltyramine(2),N-trans-caffeoyltyramine(3),[2-(3,4-dihydroxyphenyl)-2-hydroxyethyl]-3-(4-methoxyphenyl)prop-2-e-namide(4),3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acry-lamide(5),dihydro-N-trans-feruloyltyramine(6),dihydro-N-caffeoyltyramine(7),lyciumide A(8),N-3,4-dihydroxyhydrocinnamoyl tyramine(9),(2S,E)-N-[2-hydroxy-2-(4-hydroxyphe-nyl)ethyl]ferulamide(10),trans-sinapoyltyramine(11),N-trans-coumaroyloctopamine(12),trans-N-acetyl-N-feruloyl-putrescine(13),(E)-2-(4,5-dihydroxy-2-{3-[(4-hydroxyphenethyl)amino]-3-oxopropyl}phenyl)-3-(4-hydroxy-3,5-dimethoxyphenyl)-N-(4-acetamidobutyl)acry lamide(14),(1,2-trans)-N3-(4-acetamidobutyl)-1-(3,4-dihydroxyphenyl)-7-hydroxy-N2-(4-hy-droxyphenethyl)-6,8-dimethoxy-1,2-dihydronaphthalene-2,3-dicarboxamide(15),1,2-dihy-dro-6,8-dimethoxy-7-hydroxy-1-(3,4-dihydroxyphenyl)-N1,N2-bis[2-(4-hydroxyphenyl)ethyl]-2,3-naphthalene dicarboxamide(16),grossamide(17),(+)-syringaresinol(18),zhebeiresin-ol(19),scopolletin(20),isofraxidin(21),C-veratroylglycol(22),(±)-eriodictyol(23),isoquercitrin(24),quercetin(25),isovanilin(26),vanilin(27),(3,4,5-trimethoxyphenyl)meth-nol(28),5,5’-dimethoxybiphenyl-2,2’-diol(29),phthalic acid,bis-(2,5-dimethyl-hexyl)easte-r(30),dibutylphthalate(31),E-ferulic acid hexacosyl ester(32),p-hydroxyphenethyl trans-ferulate(33),1-octacosanol(34),steraric acid(35),β-sitosterol(36),lyciumin A(37).Compound 1*is a new compound,compound 28 is a new natural product,and compounds4-5,12,14,16,18-19,23,26,29,32-34,37 are isolated from this plant for the first time.Measuring the cytotoxic and inhibition of NO production activities of the above compounds by using MTT method and Griess reagent colorimetry,it was found all the compounds showed no cytotoxicity against RAW 264.7 macrophages.Compounds 3,5,8,17 and 25 showed inhibition of NO production activities with the IC500 values 8.89±0.57,17.00±1.11,6.68±0.77,14.57±2.23,17.21±0.5μg/mL,respectively.
