| Objective:Observe the effect of Dendrobium mixture on the cognitive function and intestinal flora of diabetic rats,initially explore some mechanisms of Dendrobium mixture to protect brain function,and provide a preliminary research basis for exploring the prevention and treatment of diabetic cognitive dysfunction based on the "gut-brain axis".Methods:Twenty-eight SD rats were selected and fed adaptively for 2 weeks and randomly divided into normal group(8)and model group(20).The normal group was fed with normal feed,the model group was fed with high-fat and high-sugar feed for 6 weeks,then 25 mg / kg STZ twice,and the diabetes model was replicated by intraperitoneal injection at intervals of 72 hours.11.1mmol / L is a diabetic model rat.Sixteen model rats were randomly divided into model group and Dendrobium mixture group according to blood glucose stratification.The normal group and the model group were given the same amount of saline,and the Dendrobium mixture group was given the Dendrobium mixture,and the Morries was used to test the learning and memory abilities of the rats.Fasting blood glucose was measured before collecting the material,stool was collected to detect intestinal flora 16 S rDNA,blood and tissue were collected after anesthesia,serum inflammatory factors IL-6,TNF-α,LPS were detected by ELISA,and the morphology of hippocampus and colon were observed by HE staining,the tight junction proteins Occludin and ZO-1 in the hippocampus and colon were detected by Western Blot.Results:1.The morris water maze test of rats in the diabetic model group showed that the escape latency was prolonged and the target quadrant stay time was shortened(P <0.05),indicating that learning and memory abilities were reduced,indicating that diabetic rats had cognitive dysfunction.The escape latency of the Dendrobium mixture group was shortened,and the residence time in the target quadrant was prolonged(P <0.05),indicating that the Dendrobium mixture can improve the learning and memory ability of diabetic rats.2.Serum ELISA test of the diabetic model group showed that the inflammatory factors IL-6,TNF-α and LPS increased(P <0.05),and the Dendrobium mixture group IL-6,TNF-α and LPS decreased(P <0.05).3.HE staining in the hippocampus of the diabetic model group showed that the number of cells in the CA1 area was reduced,unevenly distributed,loosely arranged,and the nucleus was constricted.Western Blot showed that the expression of the tight junction proteins Occludin and ZO-1 in the hippocampus was reduced(P <0.05).The Dendrobium mixture group hippocampus CA1 area The number of cells is relatively large,the distribution is uniform,the arrangement is regular,and the expression of the tight junction proteins Occludin and ZO-1 in the hippocampus increases(P <0.05).4.HE staining of the colon in the diabetic model group showed continuous epithelial destruction,goblet cell reduction,and more lymphocyte infiltration in the lamina propria.Western Blot showed a decrease in the expression of Occludin and ZO-1(P<0.05)in the colon tight junction protein.Colon epithelium in the Dendrobium mixture group The cells arerelatively complete,the intestinal glands are arranged relatively regularly,there is a small amount of lymphocyte infiltration in the lamina propria,and the expression of tight junction proteins Occludin and ZO-1 in the colon is increased(P <0.05).5.The diversity of intestinal microflora in diabetic rats decreased,the abundances of Thick-walled bacteria,Proteobacteria,Prevotella increased,the abundance of Bacteroides,Lactobacillus,Bifidobacterium decreased,Dendrobium mixture group,the abundance of Bacteroides,Lactobacillus,Bifidobacterium,etc.increased,while the abundance of Thick-walled Bacteria,Proteobacteria,Prevotella,etc.decreased.Conclusions:1.Dendrobium mixture can improve the learning and memory functions of diabetic rats.2.Dendrobium mixture has a protective effect on the blood-brain barrier of diabetic rats,which is related to reducing the inflammatory factors IL-6,TNF-αand LPS,and increasing the expression of tight junction proteins Occludin and ZO-1.3.The reduction of inflammatory factors by Dendrobium mixture may be related to regulating intestinal flora,increasing the expression of colon tight junction proteins Occludin and ZO-1,and protecting the intestinal barrier. |
