Objective: To explore the protective effect of Puerarin on acute liver injury in rats with sepsis and its possible mechanism.Methods: 54 healthy and clean SD rats were divided into three groups according to the random number table: sham group,sepsis group and puerarin group(18 rats in each group).Six rats in each group were randomly selected to observe the 72 h survival rate.The remaining rats in each group were randomly divided into 12 h and 24 h subgroups according to the experimental time node,six rats in each subgroup.In sham group,only the cecum was opened and flipped.CLP Group and pue group were used to establish sepsis rat model.Puerarin intervention was given in pue group at 0 h and 6 h after operation,and 80 mg / kg intraperitoneal injection was given in the other groups at the same time.According to the time node,blood and liver tissue samples were collected in each subgroup.Serum TNF-α,IL-6,NF-κ B,ALT,AST,MDA content and SOD activity in liver tissue were measured.Pathological changes of liver tissue were observed under microscope after he staining.Results:(1)serum levels of TNF-α,IL-6,NF-κ B and ALT in CLP Group and pue group were significantly higher than those in sham group(P < 0.05),and the levels of TNF-α,IL-6,NF-κ B and ALT in pue group were significantly lower than those in CLP Group(P < 0.05).Compared with sham group,the serum ast level of CLP Group and pue group increased significantly(P < 0.05);compared with CLP Group,the serum ast level of pue group at 12 hours after operation had no statistical difference(P > 0.05),and the serum ast level of pue group decreased significantly(P < 0.05)at 24 hours after operation.(2)MDA content of liver tissue: compared with sham group,MDA level of CLP Group and pue group at each time point was significantly higher(P < 0.05);compared with CLP Group,MDA level of pue group at each time point was significantly lower(P < 0.05).(3)SOD activity of liver tissue: compared with sham group,SOD activity of liver tissue in CLP Group and pue group decreased significantly(P < 0.05)at each time point;compared with CLP Group,SOD activity of liver tissue in pue group increased significantly(P < 0.05)at 12 h postoperatively,and there was no statistical difference in SOD activity of liver tissue in pue groupat 24 h postoperatively(P > 0.05).(4)Histopathological changes of liver: in sham group,the nuclei of liver were in the middle,arranged in order,without obvious changes in morphology,without inflammatory cell infiltration,and there was no significant difference at 12 h and 24 h time points;in CLP Group,the hepatocytes were not evenly arranged at 12 h,with a small number of vacuoles forming and inflammatory cell infiltration.In pue group,the hepatocytes were arranged relatively orderly in 12 h,with a small amount of vacuole degeneration,which was less pathological damage than CLP Group at the same time point.A large number of inflammatory cells infiltrated into the liver interstitium at 24 h,and the degree of injury was significantly reduced compared with the CLP group at the same time point.(5)72h survival rate: Puerarin had no effect on the 72 h survival rate of sepsis rats.
