| Chinese medicinal materials is an important part of traditional Chinese medicine.Strengthening the management of Chinese medicinal materials and ensuring the quality and safety of Chinese medicinal materials are of great significance for ensuring public health and promoting the sustained and healthy development of Chinese medicinal materials industry.How to effectively prevent sub-optimal,dyeing weight gain,adulteration and other acts of counterfeiting is a serious challenge to the quality control of traditional Chinese medicine.In particular,the use of industrial dyes for illegal dyeing seriously affects the quality and safety of traditional Chinese medicine.It is particularly urgent to establish an efficient detection method for industrial dyes.According to the results of special sampling of Chinese medicinal materials and decoction pieces published by the CFDA,19 common representative industrial dyes were selected as the research objectives.To establish a rapid and sensitive method for the determination of illegally added industrial dyes in traditional Chinese medicines,which will be applied to the quality control of traditional Chinese medicines to ensure the quality and safety of traditional Chinese medicines.In the first chapter,The present situation of illegal dyeing of Chinese medicinal materials and the toxicity of industrial dyes were systematically introduced.The research progress of industrial dye detection methods was reviewed in detail.At present,most of the reports on dye detection at home and abroad focus on food detection,but few on industrial dyes in traditional Chinese medicines.Supplementary inspection methods and approval documents for inspection items of Chinese medicinal herbs and Chinese medicinal herbs involve fewer kinds of industrial dyes in Chinese medicinal herbs,and there is no liquid phase quantitative method.The guiding principle of pigmentation method in Chinese Pharmacopoeia is not universally applicable,and the detection frequency of red dyes is the highest.Therefore,it is necessary to establish a variety of detection methods for red industrial dyes first.In the second chapter,An ultra-high performance liquid chromatography-diode array detector(UHPLC-PDA)method was developed for the simultaneous determination of a variety of illegally added red industrial dyes in Chinese medicinal materials.Different chromatographic column,different liquid mobile phase separation effect,different extraction methods,according to different industrial dyes uv spectrum,determine the quantitative detection wavelength.The results showed that 10 kinds of red industrial dyes were separated and detected at the same time in 40 min,and the detection limit of the instrument was 0.02 ~ 0.09 mg /L.This method has the advantages of high speed,high sensitivity and good reproducibility.The method was applied to the detection of 70 batches of samples.Sudan red I was detected in Dragon’s blood samples and eosin was detected in fruit of Chinese magnoliavine samples.Due to the complex matrix of Chinese medicinal materials in the detection process,the chromatographic method is easy to be interfered,affecting the qualitative and quantitative results.In order to further improve the flux,sensitivity and application range of the detection method.Secondly,it is necessary to establish a HPLC-MS method which can detect many kinds of industrial dyes at the same time.In the third chapter,The UHPLC-MS/MS method was established for the simultaneous determination of 19 illegal colouring agents in Chinese medicinal materials.The conditions of mass spectrometry and liquid chromatography were optimized.The second-level debris-ms spectrum librar was established,which provided a method basis for the quantitative and qualitative determination of dyes.The results showed that a variety of industrial dyes were separated and detected at the same time for 30 min,and the detection limit of the instrument was 0.07~2.11 μg/L.The method has high throughput,high sensitivity and good reproducibility.The method was applied to actual samples and confirmed by mass spectrometry.In the fourth chapter,the systematic summary is made,and the future of the detection methods of industrial dyes added illegally in Chinese medicinal materials is prospected. |
