Objective To study the effects of Tuber Fleeceflower Stem Decoction on synaptic protein expression and synapses development in hippocampus of depression model rats induced by CUMS.Methods 150 SD rats were randomly divided into blank control group,Ovx group,Ovx+L group,Ovx+M group,Ovx+H group and Ovx+P group,each group was further divided into 2 weeks group and 3weeks group.Except for the blank control group,the rest of the groups used separation combined with chronic unpredictable mild stress for 5 weeks to establish depression rats model.After the modeling,the blank control group and the Ovx group were given double distilled water,the Ovx+P group was given fluoxetine,the Ovx+L group,Ovx+M group and Ovx+H group were given different doses of Tuber Fleeceflower Stem Decoction for intragastric administration.The two-week group and three-week group were gavaged for 14 days and 21 days.Detect the changes in the physique of rats by the body weight.The sucrose preference test,open field test,forced swimming test were used to evaluate the depression behavior of rats.The expression of 5HT1 A receptor and synaptic protein SynapsinⅠand PSD 95 in the hippocampus tissues were detected by Western blot.Nissl staining was used to observe the damage of rat hippocampal neurons.Golgi staining was used to observe the number and morphological changes of dendritic spines in hippocampal CA1 area of rats.Results 1.The results of rat body weight show that Tuber Fleeceflower Stem Decoction can significantly increase the body weight of depression model rats.2.Behavioral test results show that Tuber Fleeceflower Stem Decoction can significantly increase the sucrose preference rate,horizontal exercise distance and vertical standing times of open-file test,reduce the immobility time of forced swimming,and significantly improve the depression behavior of rats.3.Western blot results showed that Tuber Fleeceflower Stem Decoction can significantly increasethe expression of 5-HT1 A receptor and synapsin Ⅰ 、 PSD95 protein in the hippocampus of depression model rats.4.The results of Nissl staining showed that Tuber Fleeceflower Stem Decoction can significantly increase the number of Nissl bodies in the hippocampal CA1 and CA3 regions,and have certain protective effects on neurons in depression model rats.5.Golgi staining results suggest that Tuber Fleeceflower Stem Decoction can significantly increase the number of dendritic spines in CUMS-induced depression model rats.Conclusions 1.Tuber Fleeceflower Stem Decoction has a certain therapeutic effect on CUMS-induced depression rats,which can significantly improve depression behaviors such as lack of pleasure,psychological despair and decreased voluntary activity.2.The antidepressant mechanism of Tuber Fleeceflower Stem Decoction may be related to increasing the number of Nissl bodies in hippocampal CA1 and CA3 regions of rats,repairing dendritic spinal injury in rat hippocampal CA1 regions,enhancing the function of 5-HT1 A receptors in the post-synaptic membrane and increasing the expression of synaptic protein Synapsin Ⅰ and PSD 95,thereby improving the abnormality of hippocampal synapse development and synaptic plasticity in rats.
