Objective:To investigate whether Dangua prescription can regulate the expression level of Nampt gene,and then affect the expression level of InsR,PKB mRNA and protein,improve IR and insulin sensitivity,and thus achieve the effect of regulating glycolipid metabolism,so as to provide ideas about preventing the disorder of glycolipid metabolism such as diabetes with Chinese medicine.Methods:Thirty 6-week-old male SPF SD rats were used as the research object.After 2 weeks of adaptive feeding and fasting for 8 hours,glucose tolerance test was performed to filter rats with naturally occurring blood glucose abnormalities.And the remaining rats were divided into normal group and building module group by the method of random numbers table.The normal group was fed with ordinary feed,and the building module group was given a special high-glucose and high-fat formula feed to induce the abnormal model of glycolipid metabolism.After 4 weeks of induction,the glucose tolerance test was performed again.The rats of building module group were divided into model group and Dangua prescription group by the method of random numbers table.The normal group was left untreated,while the model group was perfused with sterile water.The Dangua prescription group was administered gavage with the Dangua prescription.After 12 weeks of intervention,samples were taken.Glucose was measured by glucose test strips through the method of glucose oxidase.Used glycated hemoglobin test card to detect HbA1 c,biochemical method to measure TC,TG and LDL-C.ELISA method to detect serum INS,and caculated AUCI,HOMA-IR,HOMA-IRI.Real-time fluorescent quantitative PCR method to detect liver tissue Nampt,InsR and PKB mRNA expression level,ELISA method to detect plasma Nampt,Western blot method to detect InsR and PKB protein expression levels in liver tissue.SPSS24.0 statistical software was used for statistical analysis and processing of experimental related data.Results:1.The basic situation in each group of rats: Through the 12 weeks intervention,compared with the model group,the rats in the normal group and the Dangua prescription group were still active,with better hair color and no significant difference in the amount of food,water and urine.The weight of the normal group increased steadily.Compared with the normal group,the weight of the model group and the Dangua prescription group increased to different extents,and the model group increased more than the Dangua prescription group.2.The FBG,AUCG and HbA1 c level in each group of rats:(1)The AUCG in the model group increased compared with the normal group(P<0.05),and the level of FBG and HbA1 c increased too(P<0.05);(2)The AUCG in the Dangua prescription group decreased compared with the model group(P<0.05),the level of FBG and HbA1 c decreased(P<0.05).3.Serum TC,TG,LDL-C level in each group of rats:(1)The serum level of TC and LDL-C in the model group increased compared with the normal group(P<0.05),and therewas not found significant difference about the level of TG(P>0.05);(2)The serum TC and LDL-C levels in the Dangua prescription group decreased compared with the model group(P<0.05),and there was not found significant difference about the level of TG(P>0.05).4.Plasma Nampt,serum FINS level and AUCI in each group of rats:(1)The level of plasma Nampt increased in the model group compared with the normal group,(P<0.05),and both the level of serum FINS and AUCI increased in the model group(p<0.05);(2)The level of plasma Nampt in the Dangua prescription group decreased compared with the model group(P<0.05),and both the level of serum FINS and AUCI decreased in the Dangua prescription group(p<0.05).5.The level of HOMA-IR and HOMA-ISI in each group of rats:(1)The level of HOMA-IR increased(p<0.05)and HOMA-ISI decreased in the model group compared with the normal group(p<0.05);(2)The level of HOMA-IR decreased(p<0.05)and HOMA-ISI increased(p<0.05)in the Dangua prescription group compared with the model group.6.Expression level of Nampt,InsR,and PKB genes in liver tissue of rats in each group:(1)The expression of Nampt mRNA in the model group was increased compared with the normal group(p<0.05),and there was not found significant difference about the expression level of InsR and PKB mRNA(P>0.05);(2)The expression of Nampt mRNA was reduced in the Dangua prescription group,and there was not found significant difference about the expression level of InsR and PKB mRNA compared with the model group(P>0.05).7.Expression level of InsR and PKB protein in rat liver tissue in each group:(1)There was not found significant difference about the expression level of InsR protein in the model group compared with the normal group(P>0.05),and the expressions level of PKB protein reduced(p<0.05);(2)There was not found significant difference about the expression level of InsR protein in the Dangua prescription group compared with the model group(P>0.05),and the expressions level of PKB protein increased(p<0.05).Conclusions:1.Dangua prescription can reduce the content of TC,LDL-C,reduce fasting blood glucose and glycated hemoglobin levels,and improve insulin resistance;2.Dangua prescription may down-regulate the expression of Nampt mRNA,increase the expression of PKB protein and improve insulin sensitivity,so as to achieve the regulation of glycolipid metabolism.
