Objective: The different expression of paired-box gene 2(PAX2)was detected in human ovarian cancer cell lines HO-8910 and HO-8910 PM,as well as in serum of patients with ovarian cancer;and its relationship with the invasion and metastasis ability of ovarian cancer were analyzed by clinical data.Methods:In vitro we cultured the human ovarian serous cystadenocarcinoma cell line HO-8910 and its paired cell line HO-8910 PM.The ability of invasion and migration in HO-8910 PM and HO-8910 cell lines was compared by scratch assay and Millicell chamber artificial basement membrane invasion assay.The expression of PAX2 mRNA and PAX2 protein in HO-8910 PM and HO-8910 cell lines were detected by Real-time PCR and ELISA,separately.Meantime,the expression of PAX2 protein in serum of 27 patients with ovarian cancer was detected by ELISA,whose clinical features were different.Results:1.The number of cells in HO-8910 PM group(89.05±3.52um)that penetrated the Matrigel membrane after 24hour’ incubation were significantly higher than in HO-8910 group(63.93±0.58)(P<0.01).2.The migration distance(348.33±10.27)μm of HO-8910 PM cell line were significantly greater than HO-8910(172±25.50)μm(P<0.01),24-hours after scratching.3.The expression of PAX2 mRNA was 0.000409±0.008232 in HO-8910 cell line,which was higher than HO-8910PM(0.000276±0.000123)(P<0.01).4.ELISA assay showed that the expression level of PAX2 protein in HO-8910 group was 2315 ±0.24pg/ml,which was significantly higher than that in HO-8910 PM group(1579 ±0.37pg/ml).Correlation analysis showed that the expression level of PAX2 protein was negatively correlated with ovarian cancer cell migration(r=-0.865,P<0.05)and invasion(r=-0.681,P<0.05).5.ELISA results show,the expression of PAX2 protein in malignant ovarian tumors,benign ovarian tumors and normal ovaries were 16.75±3.50pg/ml,31.69 ±30.53pg/ml and 23.23 ±18.69 pg/ml,respectively.The expression of PAX2 protein in serum of patients with benign ovarian tumors and patients with ovarian lesion-free was significantly higher than those of patients with ovarian cancer(p< 0.05).PAX2 protein’s expression in serum of patients with recurrence and metastatic cancer was 14.2 ±3.3 pg/ml,which was significant lower than those of newly diagnosed patients(p < 0.05).6.Among the newly diagnosed patients,patients with high-grade ovarian cancer(high-grade serous cancer)were younger than patients with non-high-grade ovarian cancer(39.1 ±14.90 vs 61.0 ±9.50 years,P<0.05).The age of patients with lateral lesions was younger than those patients with bilateral lesions,and the difference was also statistically significant(58.4 ±13.50 vs.44.5 ±16.30 years,P< 0.05).However,the age has no significant difference between the pathological stage and the pathological type(P>0.05).There was also no significant difference in the expression level of PAX2 in the serum of patients on the lesions,the pathological grades and types(P> 0.05),which were newly diagnosed.Conclusion:1.PAX2 gene mRNA and its coding protein were both expressed in highly invasive ovarian cancer cell line HO-8910 PM and its parent cell line HO-8910,and the expression of PAX2 mRNA and protein in HO-8910 was significantly higher than that in HO-8910 PM.The loss-expression of PAX2 gene may play important role in the metastasis and the invasion of ovarian cancer.2.The expression of PAX2 protein in serum of patients with ovarian cancer was lower than that of patients with benign ovarian tumor,and the patients with recurrence and metastasis tumor has lower expression level of PAX2 protein in serum than that of patients with newly diagnosed ovarian cancer.The decreased expression of PAX2 may be involved in the mechanism of invasion and migration of ovarian cancer,PAX2 may be an indicator of prognosis and therapeutic targets for ovarian cancer.
