| Objective:To explore the effects of butylphthalide on the PINK1/Parkin signaling Pathway in mitophagy with celebral ischemia-reperfusion injury in rats. Methods:Healthy adult male SD rats were randomly divided into 4 groups : sham operation group, cerebral ischemia-reperfusion group, butylphthalide group, and mitophagy inhibitor group,10 rats in each group. The model of cerebral ischemia-reperfusion was established by the longa method. The effects of butylphthalide on mitophagy through PINK1/Parkin signaling Pathway with cerebral ischemia-reperfusion injury in rats were investigated by making the Longa score, measuring the cerebral infarction volume with TTC staining, assaying the brain slices with HE staining technology and assessing the expressions of PINK1、Parkin、P62 with immunohistochemistry. Results:Butylphthalide and mitophagy inhibitor reduced the Longa score, decreased cerebral infarction volume, improved the brain damage, and alleviated the expressions of PINK1、Parkin,increased the expression of P62.But there was no significant difference between the butylphthalide group and the mitophagy inhibitor group. Conclusion:Butylphthalide can improve cerebral ischemia-reperfusion injury in rats, which may related to the inhibition of mitophagy mediated by the PINK1/Parkin signaling Pathway. |
