Objective:The expressions and differences of HMGB1,Beclin1,LC3 and AR in benign prostatic hyperplasia and prostate cancer tissues were detected to explore and analyze the correlation between HMGB1 and autophagy related proteins Beclin1,LC3 and AR and blood PSA,clinical staging,grading,biochemical recurrence and clinical metabolic indexes in prostate cancer patients.So as to provide a possible basis for the occurrence,development,treatment and prognosis of prostate cancer.Methods : Collecte 40 cases with complete clinicopathological data that underwent radical prostatectomy of prostate cancer in General Hospital of Ningxia Medical University from January 2010 to August 2018 as the observation group,and 15 patients diagnosed with benign prostatic hyperplasia during the same period were randomly selected as the control group.After all the pathologies were confirmed by HE staining,the expressions of HMGB1,Beclin 1,LC3 and AR in 40 cases of prostate cancer and 15 cases of benign prostatic hyperplasia were detected by immunohistochemistry.Application of immunohistochemical staining,observed under optical microscope analysis of dyeing results,using SPSS24.0statistical software for statistical analysis,chi-square test,rank and inspection,and Fisher’s exact probability method and rank correlation analysis method,analysis of HMGB1,Beclin1,LC3 and AR in the expression of benign prostatic hyperplasia and prostate cancer tissue,and analysis of protein expression in patients with prostate cancer clinical parameters,including patient age,serum PSA,Gleason scores,TNM stage,pathological classification,etc.)the relationship between;The expression differences of HMGB1,Beclin1,LC3 and AR in benign prostatic hyperplasia and prostate cancer were tested by chi-square test.The positiveexpression of HMGB1,Beclin 1,LC3 and AR in prostate cancer and the correlation between the positive expression level and clinical data were analyzed by Spearman level correlation analysis.Kaplan-meier method was used to analyze the relationship between the expression of HMGB1,Beclin1,LC3 and AR proteins and the survival time of prostate cancer patients.The significance of survival curve was compared and analyzed by log-rank method.Statistically significant was judged by the value of 0.05.Results:(1)Localization of HMGB1,Beclin1,LC3 and AR in benign prostatic hyperpla sia and prostate cancer tissues: HMGB1 and AR were mainly expressed in the nucleus,and a few were found in the cytoplasm when the staining was strongly positive.Beclin1 and LC3 positive staining were mainly expressed in cytoplasm,and nuclei were occasionally seen when they were strongly positive.(2)The positive expression rate of HMGB1 in benign prostatic hyperplasia and prostate cancer tissues was 20.0%(3/15)and 60%(24/40),respectively(p=0.008).ThePositive expres-sion rate of Beclin1 in benign prostatic hyperplasia and prostate cancer was 46.7%(7/15)and80.0%(32/40),respectively(P=0.0481).The positive expression rate of LC3 in benign prostatic hyperplasia and prostate cancer was 66.7%(10/15)and 77.5%(31/40),respectively(P=0.411).The positive expression rate of AR in benign prostatic hyperplasia and prostate cancer tissues was 100.0%(15/15)and 72.5%(29/40),respectively(P=0.023).The positive expression rates of HMGB1,Beclin1 and AR in benign prostatic hyperplasia and prostate cancer were statistically significant,while the positive expression rates ofLC3 in benign prostatic hyperplasia and prostate cancer were not statistically significant.(3)The Spearman correlation analysis,to analysis the HMGB1,Beclin1,LC3 and AR expression in prostate cancer organizations,and between the clinical data of patients with correlation analysis,the results showed that HMGB1 was associated with clinical stage(0.523,P=0.001),pathological grade(-0.359,0.023),biochemical recurrenc(-0.358,0.023)and neurovascular invasion(-0.362,P=0.022);Beclin1 was associated withpathological grade(0.385,P=0.014),clinical stage(0.320,P=0.044),neurovascular invasion(-0.469,P=0.002)and positive incisor margin(-0.361,P=0.022)of prostate cancer patients,with statistical significance(P< 0.05).LC3 was associated with preoperative PSA of prostate cancer patients(0.380,P=0.015),which was sta tistically significant(P< 0.05).AR was correlated with the clinical stage of prostate cancer(0.381,P=0.015),with statistical significance(P < 0.05).(4)By using the rank correlation(Speqrman correlation analysis)analysis method,the correlation analysis between HMGB1,Beclin1,LC3 and AR was conducted,and it was found that The HMGB1 is related to Beclin1(0.353,P=0.025)and AR(-0.320,P=0.044)(P<05).(5)Patients of prostate cancer were followed up,the positive and negative expressions of HMGB1,LC3 and AR were not significantly correlated with the survival of prostate cancer pa tients.Positive Beclin1 expression and negative Beclin1 expression were statistically correla te d with survival in prostate cancer patients,and patients with positive Beclin1 expression had a worse prognosis than those with negative expression.Conclusion:(1)HMGB1,Beclin1 and AR may play a role in the development of prostate cancer.(2)The expression of HMGB1 and Beclin1 protein was correlated with the clinical stage,pathological grade,positive resection margin and neurovascular invasion of prostate cancer patients.The expression of AR protein is related to the clinical stage of prostate cancer patients,and HMGB1,Beclin1 and AR may be involved in the development and infiltration of prostate cancer,promoting the progress of prostate cancer.(3)HMGB1 can be used as a predictor of the recurrence of the post-operative regression of the patient.The prognosis of prostate cancer patients with positive Beclin1 expression is relativ ely worse than patients with negative Beclin1 expression,suggesting that Beclin1 expression in prostate cancer may promote the progress of prostate cancer and shorten the survival time of patients,and Beclin1 expression in prostate cancer is expected to become amolecular marker to judge the prognosis of prostate cancer.
