| Objective:To explore the expression of FOXC2 in gastric adenocarcinoma and the association with clinicopathological factors and prognostic data.Preliminary study on the effect of inhibiting FOXC2 expression on invasion and migration of gastric adenocarcinoma cells.Initially explore the role of FOXC2 in the stemness maintenance gastric adenocarcinoma.Method:(1)The expression of FOXC2 in gastric adenocarcinoma tissues was detected by immunohistochemical staining.The results of immunohistochemical staining were scored according to established rules.The chi-square test was used to analyze the correlation between FOXC2 staining results and related clinicopathological data,and the Cox proportional hazard model was established to evaluate the impact of FOXC2 on the prognosis of gastric cancer.(2)Using si RNA interference technology to inhibit the expression level of FOXC2 in gastric adenocarcinoma cells.The effect of FOXC2 supression on the biological function of gastric adenocarcinoma cells was investigated by Transwell and scratch test.(3)Flow cytometry and stem cells sphere-forming assay were used to investigate the effect of the effect of FOXC2 inhibition on the stmeness maintenance of gastric adenocarcinoma cells.Result:(1)The expression level of FOXC2 in gastric adenocarcinoma was correlated with TNM stage,differentiation degree,Laurén classification,presence or absence of recurrence and lymph node metastasis in patients with gastric adenocarcinoma(P<0.05).In the multi-factor Cox risk proportional regression model,FOXC2 was an independent risk factor for the prognosis of gastric adenocarcinoma(P<0.05).(2)In the cell function experiments,the suppression of FOXC2 expression inhibited the migration and invasion of gastric adenocarcinoma cells(P<0.05).(3)Inhibition of FOXC2 expression significantly reduced the ratio of CD44 cells in gastric adenocarcinoma cells,and the sphere formation efficiency was also significantly reduced(P<0.05),suggesting that it inhibited the stmeness maintenance of gastric adenocarcinoma.Conclusion:FOXC2 is overexpressed in gastric cancer tissues and is associated with TNM staging,differentiation,Laurén classification,recurrence and lymph node metastasis in patients with gastric adenocarcinoma.Overexpression of FOXC2 is associated with poor prognosis in patients with gastric adenocarcinoma and is an independent prognosis risk factor for of gastric adenocarcinoma.Suppression of FOXC2 expression inhibits the invasion and migration of gastric adenocarcinoma cells,reduces the subpopulation ratio of CD44 cells,and inhibits the sphere formation efficiency.It is suggested that FOXC2 is involved in the invasion and migration of gastric adenocarcinoma and is associated with the stemness maintenance of the cancer. |
