| Objective:At present,depression is the second most common disability disease after heart disease in the world,so more and more attention has been paid to the treatment of depression.Although a large number of literatures have reported on the mechanism of depression and treatment methods,there is still no definite conclusion.At present,most of the treatment of depression is mainly western medicine.Western medicine has great limitations,causing more side effects and less obvious treatment effect,but increasing the severity of depression or drug resistance.Therefore,it is very urgent to find new drugs.Depression is a kind of psychosomatic co-morbidity,which needs to be treated from multiple targets.There are many components of traditional Chinese medicine,which can be treated by multiple targets,and there are few side effects,so the treatment of depression has attracted much attention in recent years.In order to study the effect of traditional Chinese medicine on the treatment of depression,this study selected Chinese medicine Jieyuwan to study in normal Wistar rats and WKY rat models.The experiment was divided into three parts:(1)First,the biological information Pathway Studio database was used to analyze the relationship between the compounds in Jieyuwan and brain regions hippocampus,prefrontal cortex and depression symptoms;(2)Applied sucrose preference test,open field experiment and forced swimming test to detect behavior changes of Wistar rats and WKY rats before and after administration;(3)Resting state functional magnetic resonance imaging(Rs-f MRI)was used to detect functional activites changes in the hippocampus and prefrontal cortex brain regions before and after administration.Therefore,it provides more support evidence for the treatment of depression with traditional Chinese medicine Jieyuwan.Methods:(1)The relationship between the components of Jieyuwan and brain regions hippocampus,prefrontal cortex and depression symptoms was analyzed by using the bio-informatics Pathway Studio database.(2)Adult male Wistar-Kyoto(WKY)rats are endogenous depression animal models,Wistar rats of the same strain are selected as the control groups.Firstly,the behavioral experiments of sucrose preference test,open field experiment and forced swimming test and 7T Rs-f MRI were used to measure behavior changes and the functional activities changes of the hippocampus and prefrontal cortex brain regions in Wistar rats and WKY rats before administration as baseline.(3)The experimental group WKY rats were randomly divided into 3groups :model group(WKY + Na Cl),Jieyuwan group(WKY + JYW),citalopram group(WKY + CPM),and Wistar rats were used as the control group(WKY +Na Cl),and given intragastric administration for 21 days,and then used the same behavioral experiment method to detect the behavioral changes of Wistar rats and WKY rats after administration,and the functional activities changes of the hippocampus and prefrontal cortex brain areas were detected by 7T Rs-f MRI.Results:1)The bio-informatics Pathway Studio database showed that some components of the Jieyuwan can directly affect the symptoms of depression,while others indirectly treat depression by acting on bio-markers in the brain regions of the hippocampus and prefrontal cortex.2)Behavioral experiment results show that: before administration,experimental group WKY rats compared with the control group Wistar rats,it was found that WKY rats had significantly lower the degree of sucrose preference(P<0.05),and the time of floating immobility in forced swimming test increased significantly(P<0.05),while in the open field experiment,the time of exploration in the central area(P<0.001)and the total movement distance(P<0.05)were significantly reduced;After 21 days of drug treatment,compared with before administration,Jieyuwan group(WKY + JYW)and citalopram group(WKY + CPM)were found the degree of sucrose preference was significantly increased(P<0.05),the time of floating immobility in the forced swimming experiment was also significantly reduced(P<0.05),the time of exploration in the central area(P<0.001)and the total distance of movement(P<0.05)were significantly increased,but there was no statistical difference between the Jieyuwan group(WKY + JYW)and citalopram group(WKY + CPM).3)Rs-f MRI results showed that:Before administration,experimental group WKY rats compared with the control group Wistar rats found that the values of Re Ho / ALFF /f ALFF in the prefrontal cortex and hippocampus of the WKY rats were reduced(P<0.01);After 21 days of administration of Jieyuwan and citalopram,it was found that Jieyuwan did not significantly change the value of Re Ho of the hippocampus and prefrontal cortex(P>0.05),and decreased the value of ALFF of the hippocampus(P<0.05)but increased the value of f ALFF of the prefrontal cortex(P<0.05);While citalopram decreased the value of Re Ho of the prefrontal cortex(P<0.05)but there was no significant change the value of Re Ho of the hippocampal brain area(P>0.05),and decreased the value of ALFF / f ALFF of the prefrontal cortex and hippocampus(P<0.05).Conclusion:The WKY rat had depression-like behaviors such as lack of pleasure,despair,and decreased cognitive ability,and in the Rs-f MRI,the values of Re Ho / ALFF /f ALFF in the hippocampus and prefrontal cortex are reduced;Jieyuwan can effectively improve the WKY rat depression-like behavior and affect the functional activities of neurons in the hippocampus and prefrontal cortex brain regions of WKY rats.It shows that the traditional Chinese medicine Jieyuwan plays an important role in the treatment of depression,and providing more evidence for clinical treatment of depression. |
