Objective:1.To explore the gene transcription profiles of left and right colon cancer,providing the molecular genetic basis for the pathogenesis,genotyping,individualized treatment and prognosis of left and right colon cancer.2.To compare the expression of PLK3 protein in left and right colon cancer,verifying the difference of PLK3 gene expression between left and right colon cancer,and to clarify its clinical significance.Methods:1.Total RNA were extracted respectively from 6 cases of left and right colon cancer,which were used to construct library and RNA sequence by the Illumina Hiseq 400 Sequencer.Then the differentially expressed genes were clustered,and analyzed GO enriched function and KEGG enriched Pathway.2.The clinical data of 80 cases of left colon cancer and 62 cases of right colon cancer were analyzed retrospectively.It was detected the expression of PLK3 protein in left and right colon cancer by immunohistochemistry.Then the correlation was analyzed between PLK3 and P53.It was compared high expression of PLK3 in left and right colon cancer.In addition,it was discussed the impact of survival and prognosis in left and right colon cancers with the different levels of PLK3.Results:1.There are many differentially expressed genes between left and right colon cancer,involving oxidative phosphorylation,TCA cycle,AGE-RAGE signaling pathway,PI3K-Akt signaling pathway,p53 signaling pathway,etcetera.Compared with right colon cancer,there are 147 differentially expressed genes up-regulated in left colon cancer and 417 differentially expressed genes downregulated.The up-regulated genes in LCC were RGMB,DMTN,TLE2,TP53I11,PRDX2,HMGN5,etcetera.The down-regulated genes were PIK3IP1,STAT3,TGFB1,PLK3,VEGFC,BCL2A1,etcetera.2.It was shown that there was significant difference in PLK3 protein expression between left and right colon cancer(P=0.017).There was no significant correlation between PLK3 and P53 expression(P>0.05).The difference between left and right colon cancer with PLK3 protein high expression was related to sex,age,maximum diameter of the tumor,vascular thrombus and TNM stage(P<0.05).The cumulative survival rate of patients with high expression of PLK3 in left colon cancer was significantly lower than that of patients with low expression(49.48 m vs 54.34 m,P<0.05).The cumulative survival rate of patients with high expression of PLK3 in right colon cancer was significantly lower than that of patients with low expression(43.80 m vs 60.67 m,P<0.05).PLK3 is not an independent prognostic factor for patients with left and right colon cancer.Conclusions:1.There are significant differences in gene transcription profiles between left and right colon cancer,which is the molecular genetic basis of the biological characteristics,clinical efficacy,survival and prognosis of left and right colon cancer.It lays an experimental foundation and provides a theoretical basis for further exploring the pathogenesis of left and right colon cancer,realizing individualized precise treatment and improving clinical prognosis.2.It is a significant difference in the expression of PLK3 between left and right colon cancer,suggesting that PLK3 may be related to the biological characteristics of left and right colon cancer.Monitoring the PLK3 expression effectively can help to predict the invasion and metastasis of left and right colon cancer.
