Objective: The high expression of Osteopontin(OPN)and the tumor stem cell marker CD133 has been shown to play an important role in the occurrence,development,progression,invasion,metastasis and prognosis of various tumors.By studying the expression of OPN and CD133 in human non-small cell lung cancer(NSCLC)tissues,and analyzing the correlation between OPN expression and non-small cell lung cancer stem cells,it provides new ideas and directions for the clinical treatment of non-small cell lung cancer.Methods: In this experiment,immunohistochemistry was used to detect the expression of OPN and CD133 in cancer tissues and para-carcinoma tissue of 52 NSCLC patients(all of them were taken from normal tissues 5 cm away from tumor tissue and confirmed by pathology without tumor infiltration).The relationship between the clinical and pathological characteristics of OPN,CD133 and NSCLC patients and the possible relationship between the two were analyzed.Results: The expression of OPN and CD133 in NSCLC was higher than in para-carcinoma tissue,and the differences were statistically significant(P<0.05).In TNM staging,OPN was related to the T stage of the original tumor(P<0.05),while CD133 was not related to the T stage of the tumor(P>0.05);In N staging of regional lymph node,the expression in patients with lymph node metastasis(N1 + N2)was significantly higher than that in patients without lymph node metastasis(N0),and the difference was statistically significant(P<0.05);In clinical staging,the positive rates of OPN and CD133 in patients with ?a were significantly higher than those in patients with stage ?(P<0.05);The patients with these two kinds of protein which were expressed positive,the double positive patients,had higher positive rate of lymphatic metastasis than the double negative patients(P<0.05).The expression of the two proteins was not significantly related to gender,age,smoking history,tumor diameter,pathological type,and tumor differentiation in NSCLC patients,and the differences were not statistically significant(P>0.05).Correlation analysis between the two showed that there was a significant positive correlation between the expression of OPN and CD133(r = 0.491;P<0.05).Conclusion: The expression of OPN and CD133 in NSCLC cancer tissues is distinctly higher than in para-carcinoma tissue and is closely related to the condition of clinical stage and lymphatic metastasis,and have no correlation with gender,age,smoking history,pathological tissue type,tumor size and degree of differentiation.The expression of OPN and CD133 in NSCLC cancer tissues are positively correlated,that is,there is a correlation between the expression of OPN and non-small cell lung cancer stem cells,and can provide new directions and targets for the clinical treatment of non-small cell lung cancer.
