Background:Colon cancer is one of the most common malignant tumors in China and around the world.With the continuous progress of society,the gradual improvement of living conditions and the increasing westernization of people’s lifestyles and habits,the incidence,morbidity and mortality of colon cancer are on the rise.Current research reveals that SRSF3 is highly expressed in many cancers and plays an important role in tumorigenesis and development.Therefore,further study of the molecular mechanism of SRSF3 in the invasion and migration of colon cancer is of great clinical significance for the early diagnosis,targeted treatment and prediction of patient prognosis of colorectal cancer.Objective:To investigate the expression of SRSF3 in human colon cancer tissues and analyze its relationship with clinical pathological parameters of colon cancer patients and its significance.In vitro cytology experiments,the expression of SRSF3 in colon cancer cells was detected,and the effects of silencing the expression of SRSF3 on colon cancer cell proliferation,invasion,and migration were further investigated.Methods:1.The expression and clinical data of SRSF3 in 398 colon adenocarcinoma patients were extracted from TCGA database.analyze the relationship between SRSF3 expression and clinicopathological parameters in patients with colon cancer.2.In vitro cytology experiments,RT-qPCR and Western blot were used to detect the expression of SRSF3 in colon cancer cells,and small RNA interference technology was used to silence the expression of SRSF3 in colon cancer cell lines.Trace test,Transwell migration and invasion experiments were used to detect the effects of down-regulation of SRSF3 expression on cell proliferation,migration and invasion of colon cancer cells.Bioinformatics software is used for analysis the correlation between SRSF3 and CD44 expression,SRSF3 and CTTNNB1 expression.By silencing the expression of SRSF3,the expression of CTNNB1 was detected and the correlation was analyzed.The GSEA gene enrichment analysis was used to obtain signal pathways that is correlated with SRSF3 expression in colon cancer.Results:The expression of SRSF3 in colon cancer tissues was significantly higher than that in the adjacent cancer tissues(p<0.01),the expression of SRSF3 was not significantly related to the age,gender and TNM of patients,but closely related to tumor stage.The expression of SRSF3 in early colon cancer is higher than that in advanced colon cancer(p<0.05).The expression of SRSF3 in five colon cancer cells was significantly higher than that in NCM460 cells(p<0.01).Interfering with the expression of SRSF3 can inhibit the proliferation of colon cancer cells(p<0.01),and interfering with the expression of SRSF3 can also inhibit the invasion and migration of colon cancer cells(p<0.01).According to bioinformatics analysis,the expression of SRSF3 was positively correlated with that of CD44(p<0.01),The expression of SRSF3 was positively correlated with that of CTNNB1(p<0.01).Decreasing the expression of SRSF3,the expression level of CTNNB1 was also significantly inhibited(p<0.01).GSEA enrichment analysis showed that SRSF3 expression was correlated with multiple tumor signaling pathways in colon cancer.Conclusion:1.SRSF3 is highly expressed in colon cancer tissue and colon cancer cells.The expression of SRSF3 is related to tumor stage.SRSF3 can be used as a marker for early diagnosis of colon cancer.2.The mechanism of SRSF3 may be through regulating CTNNB1 to promote the proliferation,invasion and migration of colon cancer cells.
