Objective Qizhi Tongluo capsule has been on the market for many years,with good clinical effect.As a large compound composed of 26 traditional Chinese medicines,its composition is complex,and the material basis of efficacy is not clear.In order to find the material basis of efficacy in Qizhitongluo capsule,determine the real active ingredients of the capsule.In this study,according to the traditional efficacy and modern pharmacological research of each medicine taste in Qizhitongluo capsule,the capsule was disassembled for research.The main research of this study is to study eight Qi and blood tonic drugs in Qizhitongluo capsule after the prescription was disassembled.The pharmacodynamic evaluation and pharmacodynamic material basis research of these eight Qi and blood tonic drugs were carried out to explore the material basis of the effect of Qi and blood tonic drugs taste.Methods 1.Hunger,high fat and prednisolone acetate were used to prepare the model of qi deficiency and blood stasis syndrome.The related behavioral characteristics,hemorheology indexes,ET-1,C-reactive protein and no content in serum were used as the observation indexes to observe the effect of Qizhi Tongluo Capsule three groups on Qi deficiency and blood stasis syndrome rats.2.The mice with Qi deficiency syndrome were prepared by exhaustive swimming.The time of exhaustive swimming,the content of glycogen in liver,the content of urea in serum,the activity of lactate dehydrogenase and the index of thymus and spleen were used as the observation indexes to observe the effects of Qizhi Tongluo Capsule three groups on the movement and immune function of mice with Qi deficiency.3.The model of blood deficiency was made by the method of acetylphenylhydrazine combined with cyclophosphamide.The WBC,PLT,RBC,HGB,MCH and HCT in peripheral blood of mice in each group were used as the observation indexes to observe the effect of Qizhi Tongluo Capsule three groups on blood deficiency.4.Using ultra-high performance liquid chromatography,establish the UPLC characteristic map of Tonifying Qi and blood medicine group,and demarcate the common peaks of the characteristic map of different batches of Tonifying Qi and blood medicine group;then identify the ownership of the common peaks by the identification and identification of the UPLC map of single medicine,lack of flavor medicine and control under the same conditions,and determine the contribution of each flavor medicine to the fingerprint.5.Using ultra-high performance liquid chromatograph,establish the serum UPLC characteristic map of normal rats,model rats,normal rats and model rats.Through the comparison of each chromatogram,analyze the potential blood entering components of the group of Tonifying Qi and blood,and preliminarily explore the material basis of Tonifying Qi and blood in Qizhitongluo capsule.Results 1.Compared with the blank group,the rats in the model group showed "Qi deficiency and blood stasis",the indexes of Hemorheology in the model group increased(P <0.05),the levels of C-reactive protein and ET-1 increased(P < 0.05),the content of no decreased(P<0.05),and the model of qi deficiency and blood stasis was successfully reproduced.The whole blood viscosity of each group was significantly lower than that of the model group(P < 0.05),the level of C-reactive protein and ET-1 in each group was significantly lower than that of the model group(P<0.05),and the content of NO was significantly higher than that of the model group(P<0.05).The results showed that each administration group could improve the hemorheology indexes of the rats with Qi deficiency and blood stasis syndrome,and Qi Zhi Tong Luo capsule was lower than the blank group in the whole blood viscosity(low cut I)(P<0.05),but in reducing the content of C-reactive protein and ET-1,and increasing the content of NO,Qizhitongluo capsule is better than Qizhitongluo capsule,which shows that it is of great significance to play the role of Invigorating Qi and blood.2.Compared with the model group,the time of exhaustive swimming in the whole prescription group and the Invigorating Qi and blood group was significantly longer than that in the model group(P<0.01).Compared with the model group,the experimental group can effectively improve the anti fatigue ability of mice,reduce the consumption of liver glycogen(P<0.05),reduce the content of urea nitrogen in mice(P<0.05),and reduce the activity of lactate dehydrogenase(P<0.01).Compared with the blank group,the indexes of thymus and spleen in the model group were significantly reduced(P<0.05),indicating that the model was successfully constructed and the immunity of the model group was reduced.Compared with the model group,the thymus and spleen indexes of the all-prescription group and the qi-supplementing group increased significantly(P<0.01),and the viscera indexes of the qi-supplementing group did not increase as much as those of the other two groups(P<0.05).It was confirmed that the qisupplementing drug taste could relieve exercise fatigue and improve immune function.3.Compared with the blank group,WBC,PLT,RBC,HGB and MCH in the model group showed a downward trend(P<0.05 or P<0.01),while HCT showed an upward trend(P< 0.01),indicating the success of modeling.Compared with the model group,WBC,PLT,RBC,HGB and MCH were increased in the whole prescription group and the tonic qi-blood group,while HCT was decreased(P<0.05 or P<0.01).Compared with the deficiency qi-blood group,the whole prescription group and the supplementing qi-blood group significantly increased HGB and MCH and decreased HCT at the same time(P<0.05 or P<0.01).It was confirmed that the supplementing qi-blood drug taste did indeed have a blood-enriching effect in qi-qi and blood capsules,which further indicated that the supplementing qi-blood drug taste played a certain role in qi-qi and blood capsules.4.By optimizing the preparation conditions and chromatographic conditions of the sample,the characteristic UPLC map of qi-nourishing drugs in qizhi tonglu capsule was established.The chromatographic conditions were as follows: Zorbax sb-c18 column(100mm×2.1mm,1.8),gradient elution of mobile phase acetonitrile(A)-0.1% phosphoric acid water(B)(0-11 min,5%~17%A;11 ~ 17 min,17% ~ 17%A;17~25min,17%~30%A;25~36min,30%~42%A;36~46min,42%~70%A),the sampling quantity was 1μL,the detection wavelength was 210 nm,the column temperature was 25℃,the flow rate was 0.3m L·min-1,the signal acquisition time was 46 min,the similarity of 8 batches of samples were all greater than 0.98,the method was stable and reliable,and 23 common peaks were identified.Through the comparison with the UPLC map of single herb and short herb in qi-yixue group,the common peak was assigned,and the results showed that the 8 herbs of qi-yixue in qizhitongluo capsule were reflected on the common peak map.After comparison with the chromatogram of the control solution,seven components were identified: gallic acid at peak no.3,paeoniflorin at peak no.8,isoflavone glycoside at peak no.11,astragaloside at peak no.15,emodin at peak no.16,ferulic acid at peak no.17 and ginsenoside Rb1 at peak no.20.5.By comparing the blood components of model rats and blank rats,blank rats and model rats,and combining with the characteristic map of UPLC,a total of 10 blood components were detected,among which 4 were prototype components and 6 were metabolic components.Conclusion The qi-yixue group in qizhi tongluo capsule was separated from the whole prescription,and the study of its pharmacodynamic material basis has certain significance for the elucidation of the pharmacodynamic material basis of the whole prescription.Through 8 for the construction of qi and blood tonic medicine fingerprint in vitro,combined with serum medicinal chemistry,the method of blank and model rats,dosing and blank model rats into the composition of blood in the body,to explore the benefit qi and blood drug in the body’s metabolism and transformation,help stilbene vermiculite in t2 dm capsule benefit of qi and blood drug efficacy material base to clarify,for stilbene vermiculite t2 dm capsules whole party material basis reveals provide conditions.It provides scientific experimental basis for compatibility of qizhi tongluo capsules provide,at the same time provide ideas for the research on large compound after efficacy material base.
