Objective: This study was designed to investigate the clinical significance and biological function of SFTA1 P in non-small cell lung cancer(NSCLC).Methods: 1.The molecular characteristics of SFTA1 P was analyzed using NCBI and protein coding ability prediction tool(Coding Potential Assessment Tool).2.Quantitative real time polymerase chain reaction(q RT-PCR)was used to detect the expression of SFTA1 P in 62 pairs of lung cancer tissues and adjacent normal lung tissues.The association between SFTA1 P and clinical prognosis was analyzed using the Kaplan-Meier Plotter.3.Full-length sequence of SFTA1 P was obtained from NCBI.We designed and synthesized SFTA1 P overexpression plasmid and interference RNA(si RNA),which were used to construct SFTA1 P overexpression interference cell models.4.CCK-8 was used to detect the effect of SFTA1 P on cell proliferation.Transwell mothed was used to detect the effect on cell migration and invasion.Annexin V-APC/7-AAD kit and flow cytometry were used to detect the effects of SFTA1 P on apoptosis.5.Nude mice were injected subcutaneously and through tail vein with overexpressing SFTA1 P cells,and the effects of SFTA1 P on the growth and metastasis of NSCLC were observed in vivo.Results: 1.SFTA1 P is located on chromosome 10p14 and has a total length of 693 bp.CAPT found that its protein coding ability was low.2.Compared with normal lung tissues,the expression of SFTA1 P in NSCLC tissues is lower(P<0.05).Kaplan-Meier Plotter analysis found that the lower expression of SFTA1 P was significantly associated with a poor prognosis(HR = 0.63,95% CI = 0.53-0.74,P <0.01)in NSCLC.3.Compared with HBE cells,the expression of SFTA1 P was the lowest in A549 and H460 cell lines(P<0.05),and the highest in H1299 and H1975 cell lines(P<0.05).4.Compared with the control group,SFAT1 P overexpressing decreased the cell proliferation,migration and invasion ability(P <0.05),and promoted cell apoptosis in A549 and H460 cells;on the contrary,SFAT1 P interfering significantly increased the cell proliferation,migration and invasion ability,and inhibited apoptosis(P <0.05)in H1299 cells.5.Nude mice subcutaneous tumor formation and tail vein metastasis experiments showed SFTA1P-overexpressing significantly inhibited tumor growth and metastasis capacity in vivo(P <0.05).Conclusions: 1.The expression of SFTA1 P in non-small cell lung cancer tissues is significantly lower than that in normal adjacent tissues,and low expression of SFTA1 P was significantly associated with poor prognosis in NSCLC.2.SFTA1 P can inhibit the proliferation,migration and invasion of NSCLC,and promote cell apoptosis.3.SFTA1 P can inhibit the growth and metastasis of NSCLC in vivo.
