Effects Of Taurine On Formaldehyde-induced Testicular Dysfunction

Objective: To explore the effect and effect of taurine(Taurine,Tau)on testicular dysfunction induced by formaldehyde(Formaldehyde,FA),and to further explore the possible mechanism,so as to provide a new idea for the effect and mechanism of taurine on reproductive function.Methods:(1)Forty-eight sexual mature male SD rats were randomly divided into control group,taurine intervention group,formaldehyde exposure group and taurine intervention formaldehyde exposure group with 12 rats in each group.Taurine was given intragastrically [100mg/(kg.d)],formaldehyde was given intraperitoneal injection [10mg/(kg.2d)],normal control group was given normal saline intragastric administration and intraperitoneal injection for 30 days,(2)The weight of SD rats was compared between groups,and the organ coefficients of testicular tissue were removed after operation.(3)The blood of SD rats was drawn,and the serum was taken after centrifugation,and the serum hormones such as testosterone(testosterone),luteinizing hormone(LH,Luteinizing hormone),follicle stimulating hormone(FSH,Follicle stimulating hormone))and gonadotropin releasing hormone(Gn RH,)were detected by ELISA method.The level of Gonadotropin releasing hormone)and the levels of serum superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)were detected by colorimetric method.(4)The testicular tissue on one side was taken for HE staining to observe the morphological and pathological changes,TUNEL to detect apoptosis,and immunohistochemical(IHC)to detect the expression of DRP1,LC3 and Becline1 antibodies.(5)The protein expression of Bcl-2 and Bax in the testicular tissue of the other side was detected by Western blote technique,and the superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)inthe testicular tissue were detected by colorimetric method.Results:(1)There was no significant difference in body weight,testicular weight and testicular organ coefficient among the three groups(p≤0.05).(2)The testicular spermatogenic cells decreased,the seminiferous tubule was disordered and the structure was loose in the formaldehyde group.After taurine intervention,it basically returned to normal.No obvious abnormality was found in the taurine intervention group compared with the normal control group.The results of TUNEL detection showed that the apoptosis of cells in the formaldehyde group was significantly higher than that in the control group,and the apoptosis in the formaldehyde group was significantly better than that in the formaldehyde group after the intervention of taurine.The results of immunohistochemistry showed that the expression of DRP1,LC3 and Becline1 in formaldehyde group was significantly higher than that in control group.After the intervention of taurine,the expression of antibody in formaldehyde group was lower than that in formaldehyde group.(3)The serum hormone levels of testosterone,luteinizing hormone(LH,Luteinizing hormone)and follicle stimulating hormone(FSH,Follicle stimulating hormone))in formaldehyde group were significantly lower than those in formaldehyde group(P ≤ 0.05).The level of gonadotropin-releasing hormone(Gn RH,Gonadotropin releasing hormone))increased significantly(P ≤0.01),and the results were statistically significant.(4)The(SOD)level of superoxide dismutase in the formalin group was significantly lower than that in the control group,and higher than that in the formaldehyde group(P≤0.05).The serum hormone level in the taurine group was significantly higher than that in the formaldehyde group(P≤0.05),and the superoxide dismutase(SOD)level in the formalin group was significantly lower than that in the control group(P ≤ 0.05).The level of malondialdehyde((MDA)),glutathione peroxidase(GSH-PX),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)in taurine group were significantly higher than those in formaldehyde group(P≤0.05)and formaldehyde group(P≤0.01),and the level of malondialdehyde(MDA)and glutathione peroxidase(GSH)in taurine group were significantly higher than those in formaldehyde group(P≤0.01).(5)After taurine intervention,the level of apoptotic protein Bax in formaldehyde group increased significantly,which basically reached the normal level,and the level of superoxide dismutase(SOD)in tissue homogenate in formaldehyde group was significantly lower than that in control group and higher than that in formaldehyde group after taurine intervention.The level of malondialdehyde((MDA)),glutathione peroxidase(GSH-PX),malondialdehyde(MDA)and glutathione peroxidase(GSH-PX)in taurine group were significantly higher than those in formaldehyde group(P≤0.05),and the level of glutathione peroxidase(GSH)in taurine group was significantly higher than that in formaldehyde group(P ≤ 0.01).Conclusion:Formaldehyde has reproductive toxicity on male sexually mature rats,and taurine can improve the male reproductive toxicity induced by formaldehyde,which can provide a theoretical basis for the treatment of male reproductive toxicity.
