Objective: Through the establishment of insulin induced vascular smooth muscle cell proliferation and migration of the model,the relationship of decoction of shu xin and the key link of on plaque,discusses decoction of shu xin through regulating the classic signaling pathways and related proteins induced by high insulin action mechanism of vascular smooth muscle cell proliferation and migration,provide a certain basis for prevention and treatment of coronary heart disease with TCMMethods: The effect of freeze-dried powder of shuxin drinking water extract on the inhibition of insulin-induced A7R5 vascular smooth muscle cells was detected by cck-8 cell proliferation assay.Wound-healing experiment was conducted to detect the inhibitory effect of shuxinyin on A7R5 cell migration in vitro.Western blot analysis of the effects of shuxinyin on the expression of related proteins in MAPK,AKT and Jak2/STAT3 signaling pathways in A7R5 vascular smooth muscle cells.Results: The results of cell proliferation and Wound-healing showed that,compared with the blank control group,high concentration of insulin could significantly induce the proliferation and migration of vascular smooth muscle cells(P < 0.05).Compared with the model group,shuxinyin at different concentrations could inhibit the effect of insulin on cell proliferation and migration,which was statistically significant(P < 0.05).Western Blot results showed that in the MAPK signaling pathway,compared with the blank control group,high concentration of insulin showed a tendency to activate ERK and inhibit the expression of P38 MAPK protein molecule.Compared with the model group,the shuxin decoction group showed a tendency to inhibit the expression of ERK molecules in a concentration-dependent manner,and the shuxin decoction group at concentrations of 0.3mg/ml and 1mg/ml could significantly inhibit the phosphorylation and had statistical significance(P < 0.05).Compared with the model group,shu xin Yin group with different concentrations showed a tendency to promote the expression of P38 MAPK molecule.In the expression of Akt signal,compared with the blank control group,high concentration of insulin activated the expression of Akt protein(P < 0.05).Compared with the model group,shuxinyin group at various concentrations showed a tendency to inhibit the expression of Akt molecule in a concentration-dependent manner.In the Jak2/STAT3 signaling pathway,compared with the blank control group,high insulin concentration showed a tendency to activate the expression of Jak2,STAT3tyr705 and STAT3ser727 protein molecules.Compared with the model group,shuxin decoction of different concentrations showed a tendency to inhibit the expression of Jak2 and STAT3tyr705 proteins,and shuxin decoction of 0.3mg/ml had a statistically significant inhibitory effect on the protein of STAT3ser727 protein molecule(P < 0.05).Conclusion: Shuxin decoction can inhibit the proliferation and migration of vascular smooth muscle cells induced by high concentration of insulin,which is related to the inhibition of ERK and STAT3ser727 protein molecules,but not related to the phosphorylation of Akt,P38,JAK and STAT3tyr705 protein molecules.
