Effects Of "Qin’s Xiao Bi Cataplasm" On The Expressions Of MMPs In Animals With Knee Osteoarthritis And Its Mechanism Of Action | | Posted on:2020-02-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:T T Lin | Full Text:PDF | | GTID:2404330647456129 | Subject:Traditional Chinese Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: The different animal models were established to evaluate the efficacy of “Qin’s Xiao bi cataplasm” and its mechanism was explored.Method: 1.The mouse model of knee osteoarthritis was established,and each group of animals was treated with corresponding drugs.HE staining was used to observe the degree of cartilage injury in each group,and ELISA method was used to detect the expressions of MMP-3 and CTX-Ⅱ in serum of each group.The above two methods were used to evaluate the efficacy of the alcohol extract of “Qin’s Xiao bi cataplasm”.2.The initial viscosity,multi-area viscosity,viscosity at fixed temperature condition of “Qin’s Xiao bi cataplasm” were detected.These properties were combined with sensory scores to calculate its comprehensive scores.3.The hot plate method and acetic acid method were used to establish the pain model.The swelling model was established by xylene acute ear swelling method.Each group was given corresponding drugs to verify the analgesic and anti-inflammatory effects of “Qin’s Xiao bi cataplasm”.4.A rat model of knee osteoarthritis was established,and the animals in each group were given corresponding drugs.Mankin score was used to evaluate the degree of animal cartilage injury.The expressions of IL-1β,MMP-1,MMP-3,MMP-13 and CTX-Ⅱ in serum of rats were detected by ELISA.The expressions of MMP-3 in articular cartilage were measured by immunohistochemistry and western blot.Small animal ultrasound was used to evaluate the knee cavity changes.All the methods were used to evaluate the efficacy of “Qin’s Xiao bi cataplasm”.Results: 1.Compared with the model group,the contents of MMP-3 and CTX-Ⅱ in the treatment group were significantly reduced(P<0.01).Compared with the positive group,the contents of MMP-3 in the high-dose group and the low-dose group of “Qin’s Xiao bi cataplasm” alcohol extract were significantly reduced(P<0.01).2.The comprehensive score of “Qin’s Xiao bi cataplasm” was 80,and the preparation process was stable;3.Compared with the model group,the high-dose group of “Qin’s Xiao bi cataplasm” could significantly extend the pain threshold of mice after administration for 1h and 3h(P<0.05).The pain torsion response and ear swelling degree in mice was reduced in all treatment groups(P<0.01),and the effect of “Qin’s Xiao bi cataplasm” on extending the pain threshold of mice and reducing ear swelling of mice in the high-dose group was better than that in the positive group(P<0.05).4.Compared with the model group,the serum levels of MMP-3,MMP-13 and CTX-Ⅱ were significantly reduced in each treatment group and high-dose group and positive control group can significantly inhibit the MMP-1(P<0.05).Except the low-dose group,IL-1β contents in treatment groups were significantly decreased(P<0.05).The high-dose group was better than the positive group in reducing the IL-1β expressions(P<0.05).The results of ultrasound examination showed that the soft tissue swelling degree of the knee joint was significantly reduced in all treatment groups except the low-dose group(P<0.01).Immunohistochemical results showed that the expressions of MMP-3 in the articular cartilage of the knee was significantly decreased in all treatment groups except the low-dose group(P<0.05).WB results showed that the expressions of MMP-3 in knee cartilage of the mice in the high-dose group and positive group was significantly reduced(P<0.01).Mankin score showed that,compared with the model control group,the Mankin score was significantly reduced in other treatment groups except for the low-dose group,and the difference was statistically significant(P<0.05).Conclusions: “Qin’s Xiao bi cataplasm” has a good analgesic and anti-inflammatory effect and can decrease the expressions of MMPs,CTX-Ⅱ and IL-1β in knee osteoarthritis model animals. | | Keywords/Search Tags: | Knee osteoarthritis, Qin’s Xiao bi cataplasm, Analgesic effect, Anti-inflammatory effect, MMPs, CTX-Ⅱ, IL-1β, Ultrasound | PDF Full Text Request | Related items |
