Cucurbitacin B Reduce Lipopolysaccharide-induced Acute Kidney Injury By Down-regulating Monocyte Chemoattractant Protein 1and High Mobility Group Box 1 Protein

Objective: Here,we explore Cucurbitacin B whether has a protective effect on septic acute kidney injury and its mechanism.Materials and Methods: 25 C57BL/6 mice were equally divided into five groups,which were control group,LPS group,LPS + Cu B 0.2 mg/kg group,LPS + Cu B 1 mg/kg group,LPS + Cu B 5 mg/kg group.In Cucurbitacin B pretreatment groups,we fed the mice with Cu B at different doses for 5 days,as described above.Then,we injected 5mg/kg LPS intraperitoneally into mice except the control group,after 20 hours all the mice were killed.Serum urea nitrogen and creatinine were detected using kits.Kidney injury molecule 1 and Neutrophil gelatinaseassociated lipocalin expression levels in kidney tissue were detected by q PCR.Renal pathological changes were observed by HE staining and PAS staining.The expression of monocyte chemoattractant protein 1 and high mobility group box 1 in renal tissue were detected by q PCR.Renal monocyte chemoattractant protein 1 expression in renal tissue were detected by immunohistochemical.Serum cytokine interleukin-10,Interleukin-1β and tumor necrosis factor-α levels were detected by Elisa.Results:1.Compared with the control group,the renal tubules in the LPS group were damaged,at the same time,the serum urea nitrogen increased,(P <0.05).There was no obvious difference in serum creatinine.In the LPS group,m RNA expression of NGAL and KIM-1 in renal tissue significantly were elevated,(P <0.05).2.In cucurbitacin 0.2 mg/kg and 1 mg/kg pretreatment groups,the renal tubular injury score was lower than that in the LPS group.But there was no obvious difference between LPS + Cu B 5 mg/kg group and LPS group.3.Compared with LPS group,the urea nitrogen of cucurbitacin B pretreatment group was significantly decreased(P<0.05),and the m RNA expression levels of KIM-1 and NGAL were significantly reduced(P<0.05).4.The protein expression of MCP-1 in kidney tissue of LPS group was up-regulated(P<0.05),and the m RNA expression levels of HMGB1 and MCP-1 were increased,(P<0.05).5.Compared with the LPS group,the cucurbitacin B pretreatment group had lower m RNA expression levels of HMGB1 and MCP-1(P<0.05),and lower protein levels of MCP-1 in kidney tissue(P<0.05),but serum cytokine IL-10 levels increased(P<0.05).Conclusions: 1.Cucurbitacin B pretreatment can reduce acute kidney injury induced by lipopolysaccharide 2.Cucurbitacin B reduces acute kidney injury induced by lipopolysaccharide via down-regulating monocyte chemoattractant protein 1,high mobility group box1 protein in renal tis sue and up-regulating the level of serum IL-10.
