| 1.ObjectiveTo explore the mechanism of the effect of oral anti-shock mixture on intestinal injury on the basis of early anti-shock rehydration in severely scalded rats,and to provide a theoretical basis for the clinical application of traditional Chinese anti-shock therapy.2.MethodsWeight measurement:Record the weight of experimental rats in each group,calculate the fluid rehydration volume according to the Parkland fluid rehydration formula,and calculate the combined anti-shock dose of the Chinese medicine in our hospital.Anesthesia:Prepare 10%chloral hydrate beforehand,and directly anesthetize the rat’s abdominal cavity according to 3m L:1000g.Shaving:After the rats are completely unable to move independently,after the anesthesia is effective,remove the hair on the back of the rats until the germinal layer of the rats,and clean the rat’s surface at the same time.Tail vein:One venous tube.Scald:Keep the constant temperature hot water tank at 98°C,directly put the back of the rat in hot water for 12 seconds,and form a pale and swollen wound of30%deepⅡ-Ⅲand TBSA.Rehydration:The tail vein of a rat after scald was quickly connected to a micropump,and at the same time,it was measured according to 1.5ml/kg.Rats in the sham injury group were not treated with resuscitation fluid.Part Ⅰ:Effect of Chinese medicine anti-shock mixture on serum Na~+and TNF-α,IL-10,IL-18 in severely scald ratsThe serum of the abdominal aorta of the experimental group rats was extracted,and the serum Na~+value and the values of TNF-α,IL-10,and IL-18 were measured in each group.The detection of three inflammatory factors was performed strictly in accordance with the ELISA reagent instructions.Statistical significance was analyzed based on the measured data.Part Ⅱ:Effect of Chinese medicine anti-shock mixture on intestinal tissue MDA and SOD in severely scald ratsThaw the intestinal tissue,mash the tissue,make intestinal slurry,centrifuge the sample supernatant,and compare according to the propylene glycol MDA reagent instruction manual and the superoxide dismutase SOD reagent instruction manual to detect the MDA content and SOD value.Analysis of statistical significance.Part Ⅲ:Effect of Chinese medicine anti-shock mixture on p38 MAPK of intestinal tissue in severely scald ratsTake refrigerated intestinal tissues,make them into test objects,and measure the value of p-p38 MAPK/p38 MAPK in rat intestinal tissues by reagents(western blotting)to understand the phosphorylation level of intestinal cells.Data analysis was statistically significant.3.ResultsPart ⅠSerum sodium:compared with the sham group(137.75±1.12),the serum sodium value of LR group(130.90±1.56)was significantly lower(P<0.01),and the rats developed hyponatremia,while the serum sodium value of HS600 group(145.70±0.87)and TCM mixture+HS600 group(144.69±1.52)were significantly higher(P<0.01),but the serum sodium value of both groups was basically within the normal range.Compared with the LR group,serum sodium in the HS600 group and the TCM mixture+HS600 group were significantly increased,but the differences were statistically significant within the range of normal serum sodium values(P<0.01).Compared with the HS600 group,the serum sodium value of the TCM mixture+HS600 group was reduced,but there was no statistical significance(P>0.05).Inflammatory factor TNF-α:there were significantly higher in TNF-αvalues of LR group(12.23±0.95),HS600 group(10.39±1.29)and TCM mixture+HS600 group(9.26±0.68)than in sham group(6.61±0.94)(P<0.01).Compared with LR group,TNF-αvalues of HS600 group and TCM mixture+HS600 group were significantly decreased(P<0.01).Compared with HS600 group,the TNF-value of the TCM mixture+HS600 group was significantly reduced(P<0.05).Inflammatory factor IL-18:compared with the sham group(161.94±10.74),the IL-18 value of LR group(237.85±15.04),HS600 group(206.95±13.38)and TCM mixture+HS600 group(182.16±24.76)were significantly increased(P<0.01 or P<0.05).Compared with LR group,IL-18 values of HS600 group and HS600 group were significantly decreased(P<0.01).Compared with HS600 group,the IL-18 value of the TCM mixture+HS600 group were significantly lower(P<0.01).Inflammatory factor IL-10:compared with the sham group(8.08±0.83),the IL-10 values of LR group(13.46±1.06),HS600 group(14.91±0.95)and TCM mixture+HS600 group(16.14±0.97)were significantly increased(P<0.01).Compared with LR group,the IL-10 values of HS600 group and TCM mixture+HS600 group increased significantly(P<0.01).Compared with HS600 group,the IL-10 value of the TCM mixture+HS600 group was significantly increased(P<0.05).Part ⅡMOD value of intestinal tissue:compared with MDA value of the sham group(0.46±0.02),MDA values of LR group(0.74±0.05),HS600 group(0.66±0.01)and TCM mixture+HS600 group(0.56±0.01)were significantly increased.Compared with LR group,the HS600 group and the TCM mixture+HS600 group both had significant decreases,among which the decrease was more significant in the TCM mixture+HS600 group,with statistically significant differences(P<0.01).SOD value in intestinal tissue:compared with SOD value in the sham group(422.88±14.89),SOD values in LR group(335.13±13.32),HS600 group(363.55±9.17)and TCM mixture+HS600 group(376.19±9.04)were significantly reduced.Compared with LR group,HS600 group and TCM mixture+HS600 group increased significantly,among which,the increase of TCM mixture+HS600 group was more significant,and the difference was statistically significant(P<0.01 or P<0.05).Part ⅢIntestinal tissue p-p38 MAPK/p38 MAPK values:the values of p-p38 MAPK/P38 MAPK in intestinal tissue were significantly increased in LR group(0.770±0.015),HS600 group(0.544±0.015)and TCM mixture+HS600 group(0.510±0.017)compared with that of the sham group(0.457±0.018)(P<0.01).Compared with LR group,p-p38 MAPK/p38 MAPK values of HS600 group and TCM mixture+HS600 group were significantly decreased(P<0.01).However,the activity of p38 MAPK in TCM mixture+HS600 group was significantly lower than that in HS600 group(P<0.05).4.ConclusionIn the liquid resuscitation of rats with severe scald,the addition of Chinese medicine anti-shock mixture hypertonic salt solution could effectively reduce the early intestinal tissue damage of rats with severe scald. |
