| Eucommiae cortex(Duzhong)is the dried bark of Eucommia ulmoides Oliv..It has many pharmacological effects including tonifying liver and kidney,strengthening bone and fetus,etc.Duzhong is widely used in clinic.But in the monograph of Eucommiae cortex recorded in China pharmacopoeia(2015 edition,Volume I),there is no TLC identification method.Eucommiae folium is the dried leaf of Eucommia ulmoides Oliv..Modern studies have shown that the chemical constituents and pharmacological effects of Eucommiae cortex and Eucommiae folium are similar.Eucommiae folium was first recorded in China Pharmacopoeia in 2005,using chlorogenic acid as the qualitative and quantitative marker.Dianüjin Wan is a compound preparation commonly used for gynecology,which consists of 36 crude drugs including Eucommiae cortex.And it has the effect of nourishing blood and kidney and regulating menstruation.In the current quality standard of Dianüjin Wan,only the content of paeoniflorin is controlled.In order to improve the quality standards of Eucommiae folium,Eucommiae cortex and Dianüjin Wan and ensure the safety of their clinical application,the studies of quality evaluation methods was carried out.1.Improvement of the quality standard of Eucommiae foliumA chemical components analysis of Eucommiae cortex and Eucommiae folium was carried out by using UPLC-Q-TOF MS method,and 33 chemical constituents were identified.The results indicated that although the main chemical composition types of Eucommiae folium and Eucommiae cortex were similar,which mainly contain lignans,iridoid,flavonoids and so on,the relative contents of these chemical components in Eucommiae folium and Eucommiae cortex are quite different.Both qualitative method by TLC and quantitative method by HPLC were established and validated for quality evaluation of Eucommiae folium.Asperuloside(ASP),chlorogenic acid(CA),astragalin(AST)and isoquercitrin(ISO)were used as the chemical markers in TLC identification method.HPLC quantitative methods for the determinations of ASP was established.In order to compare the difference of the contents between young and old leaves of Eucommia ulmoides Oliv.,a HPLC-DAD method for simultaneous determination of 9 components including aucubin,geniposidic acid,asperuloside,chlorogenic acid,rutin,quercetin 3-O-sambubioside,isoquercitrin,kaempferol-3-O-rutinoside and astragalin was established.The results showed that the difference of the relative content of main chemical composition between young and old leaves of Eucommia ulmoides Oliv.was great.The content of AU,GPA,ASP and CA in younger leaves of Eucommia ulmodies Oliv.was significantly higher than that old leaves.2.Improvement of the quality standard of Eucommiae cortexUsing pinoresinol diglucoside(PDG)and Geniposidic acid(GPA)as the chemical markers,a new TLC method of Eucommiae cortex was established,which supplying a gap for its quality standard.3.improvement of the quality standard of Dianüjin WanThe TLC identification method and the HPLC content determination method of Paeoniae radix alba and Rhizoma Cyperi of Dianüjin Wan were studied by using paeoniflorin and alpha-cyperone as the chemical markers respectively.The methodology and robustness of the methods were investigated as well.The results showed that these methods were simple,rapid,reliable,accurate and without negative interference. |
