| 【Objective】 0.01% Hypochlorous acid is a common antimicrobial agent with low toxicity, strong oxidation, strong sterilization ability, and low drug resistance. It is widely used in daily life and skin and mucous membrane disinfection. There are few studies on extinction. This study proved in vitro experiments that 0.01% hypochlorous acid can kill common pathogens(including bacteria, fungi and viruses)of skin and mucous membranes, and compared with the common iodine skin disinfectants and 75% ethanol disinfectants commonly used in hospitals to explore the killing effect of 0.01%hypochlorous acid on pathogens. The purpose is to provide experimental basis for the clinical search for an ideal,low-resistant antimicrobial agent.【Methods】 1.Dilute the standard bacterial strains, including Staphylococcus aureus,Escherichia coli,Hemolytic streptococcus,Pseudomonas aeruginosa, with sterile physiological saline to prepare a bacterial suspension with a concentration of 1 x 108 cfu/ml ~ 5 x 108 cfu/ml bacteria suspension. The prepared bacterial suspension and the 0.01% hypochlorous acid solution, anal iodine skin disinfectant, and 75% ethanol disinfectant have different effects.Draw, culture and count, determine the number of viable bacteria according to the live bacteria culture counting method, and calculate the average viable bacteria concentration of each group.Use the log kill value and kill rate to evaluate the sterilization effect.2.Dilute the standard fungal strains, including Trichophyton rubrum,Malassezia and Candida albicans with sterile saline,to prepare a bacterial suspension with a concentration of 1 x 108 cfu/ml ~ 5 x 108 cfu/ml fungal suspension. The prepared bacterial suspension and the 0.01% hypochlorous acid solution, anal iodine skin disinfectant, and 75% ethanol disinfectant have different effects.Draw, culture and count, determine the number of viable bacteria according to the live bacteria culture counting method, and calculate the average viable bacteria concentration of each group.Use the log kill value and kill rate to evaluate the sterilization effect.3.Adopt enterovirus type 71 infection of African green monkey kidney cell(Vero cell)model and its cell infection technology, respectively, with 0.01% hypochlorous acid solution, anal iodine skin disinfectant and 75% ethanol disinfectant for different time.Using the endpoint dilution method to calculate the virus infection titer.Use the log kill value and kill rate to evaluate the virus killing effect.【Results】 1.The killing rate of bacteria by 0.01% hypochlorous acid is: E.coli 99.999%,Staphylococcus aureus 99.999%,Pseudomonas aeruginosa>99.9999%,hemolytic streptococci>99.9999%,and 75% ethanol disinfectant. There was no obvious difference in the sterilization rate of Aneriod skin disinfectant.2.The killing rate of 0.01% hypochlorous acid on fungi are: Candida albicans 99.999%,Malassezia 99.9%,Trichophyton rubrum 99.999%,and 75% ethanol disinfectant and iodine skin disinfectant. There was no obvious difference in bacterial rate.3.The killing rate of 0.01% hypochlorous acid on enterovirus type 71 was 99.99%,and there was no obvious difference from the 75% ethanol disinfectant and anal iodine skin disinfectant to kill the virus.【Conclusion】 1.0.01% hypochlorous acid in vitro for 5 minutes,it is common to skin and mucous membrane Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa, hemolytic streptococcus,Candida albicans,Malassezia,Trichophyton rubrum And pathogenic microorganisms such as enterovirus type 71 have a good killing experiment.2.In comparison with 0.01% hypochlorous acid and the common iodine skin disinfectant and 75% ethanol disinfectant commonly used in hospitals, the study found that there is no significant difference in vitro killing experiment on the above-mentioned common pathogenic microorganisms of skin and mucosa. |
