| Background Clear cell renal cell carcinoma(cc RCC)is one of the most common malignant tumors of the urinary system.However,the mechanisms underlying the development and progression of clear cell renal cell carcinoma remain largely unknown.Recent studies have shown that circ RNA are dysregulated in human tumors and participates in the occurrence and development of many kinds of tumors.However,little is known about the role of circ RNA in the pathogenesis and development of clear cell renal cell carcinoma.Methods 3 pairs of human clear cell renal cell carcinoma tissues and matched paracancerous tissues were sequenced by high-throughput circular RNA,and the dysregulated circ RNA was screened by bioinformatics analysis.Then,the m RNA,mi RNA and circ RNA sequencing data of clear cell renal cell carcinoma were downloaded from The Cancer Genome Atlas database and Gene Expression Omnibus database.Based on the above sequencing data,we screened out the differentially expressed circ RNA,mi RNA and m RNA,and combined with the prediction website to build a competitive endogenous RNA network.Finally,we use LASSO regression,multivariate COX regression and other statistical methods to explore the relationship between the network and the prognosis of the disease Results First of all,through the sequencing of clinical samples,We obtained 509 circ RNAs that were differentially expressed between clear cell renal cell carcinoma and adjacent tissues.Secondly,a competing endogenous RNA network with 14 circ RNA,36 mi RNA and 314 m RNA is constructed.This network may promote tumorigenesis and development by affecting WNT pathway,kidney development and uroepithelial system development.Finally,through multivariate COX regression and LASSO algorithm,we found that the downstream m RNA regulated by the network pathway of 11 circ RNA was related to tumor prognosis and could well predict the 5-year survival rate of tumor patients.Conclusion There are a large number of differences in the expression of circ RNA in renal clear cell carcinoma and paired paracancerous tissues,which may promote tumorigenesis and development through competing endogenous RNA networks including circ RNA/mi RNA/m RNA,but its value needs to be further verified. |
