BackgroundMagnetic(Fe3O4)nanoparticles(NPs)can be targeted to intented sites by external magnetic fields.However,Fe3O4 NPs used for biomedical applications are usually treated with surface modifications.The modification can increase the chemical stability and improve the biocompatibility of the Fe3O4 NPs.A lot of materials have been used to modify Fe3O4 NPs,such as silica,carbon,Ag,PLGA,etc.Carboxymethyl-chitosan is a water-soluble chitin derivative.The structure of cmCHI contains the amino and carboxyl groups.The carboxyl groups on molecular can firmly combined with hydrogen bond on Fe3O4 NPs.And amino can be connected with drugs and other ligands.Floate is emerged as a active targeting ligand for selective delivery of curcumin to cancer positions.It is reported that the folate receptors over-express in many cancer tissues.AimHere we developed Fe3O4 NPs coated with Carboxymethyl chitosan(cmCHI)and introduce flote(FA)cross-linked cmCHI(FA/cmCHI/Fe3O4 NPs)as a delivery system,researching the mechanisms of this delivery system.Methods1.We developed Fe3O4 NPs coated with Carboxymethyl chitosan(cmCHI)by chemical crosslinking and then with floate.Scanning electron microscopy(SEM),X-ray diffraction(XRDand fourier transform infrared spectroscopy(FTIR)were used to measure morphology,physical and chemical structure of the nanoparticles.2.To study the effects of FA/cmCHI/Fe3O4 NPs on cell apoptosis and necrosis,we choose human lung cancer cell line A549 cells without flote receptors and cervical cancer line HeLa cells with flote receptors as model cells.Results1.A series of physical and chemical characterization showed that FA/cmCHI/Fe3O4 delivery system was synthesized successfully.2.The experiments showed that only when the concentration of FA/cmCHI/Fe304 Nps is relatively high that they would have some effects on cell apoptosis and necrosis.ConclusionThis study revealed that the loading capacity of FA/cmCHI/Fe3O4 Nps is actually great and enough medicines can be brought to the intented sites.And the drug released with a certain way and a certian rate.And the medicines can be maintained in the targeting areas for a period of time in a effective concentration...
