Pyrrolidones are kind of alkaloid widely existing in nature and synthetic drugs which has a variety of biological activity and important applications in medicine,food,chemistry and other fields.Its amide ring structure in the basic skeleton is stability and strong plasticity,so variety pyrrolidone derivatives can be acquired.Therefore its plenitudinous development and application is significant.In this paper we studied the synthesis and biological activity of novel pyrrolidone compounds from the anti dementia drug Phenchlobenpyrrone refering to the data in previous research achievements.Through substitution,addition,reduction,condensation,cyclization and other che mical synthetic method,a series of products were synthesized with Phenchlobe npyrrone or substituted benzaldehyde derivatives as raw materials Including 4,5-s ingle aromatic ring substituted pyrrolidones,17 Phenchlobenpyrrone derivatives,a product with new parallel loop,and 3 reducteded products.Their structures wer e identificated by MS and NMR spectra.in addition,The reseach indicated that t he copper iodide system is capable of breaking N-chain in pyrrolidines.Previous works showed some pyrrolidones expressed blood toxicity: hemolysis.Consequently hemolytic degree of the products were investigated with spectropho tometry,gained the hemolytic results of more than 40 different compounds.After analysising,the relationships between the the structures of the compounds and h emolysis were cleared.Studied on the antibacterial activity test of pyrrolidones,found that some of the compounds had considerable antibacterial activity to bacterial and fungal.The result proved that this kind of alkaloid have certain value of research and application to antibacterial activity.
