Bidrectional-fermentation is called the medicinal fungal new type(bidirectional)solid-state fermentation engineering,that the active ingredients of Chinese herbal medicines are used as medicinal substance to replace the traditional fungal culture(wheat bran,bagasse and the other agricultural products).The potency substance in the fermentation process not only provides nutrients such as carbonitride which fungal requires,the enzyme which is produced by the fungus can also change the component of herbs,achieve better efficacy,produce new nature and flavor,function 5 and reduce the toxicity of raw herbs.Bidirectional fermentation technology can be applied without separating wtih medicinal fungal’s secondary metabolites and work together to make better medicinal effect.Trametes robiniophila Murr,one of the most successful medicinal fungi·Huai Qi fungal substance has achieved a good clinical effect.Radix Isatidls is commonly applied in the field of detoxification,cooling pharynx and the other effects.It is widely used in traditional Chinese medicine clinical treatment of various diseases.Besides,its chemical composition of basic research has been mature.This study will build the Trametes robiniophila Murr and Radix Isatidls fermentation system,conduct bi-fermentation to optimize its optimum fermentation conditions,to determine the end of fermentation period,and through undertaking the study of the chemical composition and pharmacodynamic to determine The quality of this new medicine,fungal substance Quality Control Standard to provide scientific basis.On this basis,we begin to undertaking the mechanism of bi-fermentation in order to get a deeper understanding of the interaction and variation between the fungal substance and the herbs.The methods and results of this study are as follows:(1)Trametes robiniophila Murr is inoculated in Radix Isatidls herb.consumption rate and polysaccharide content change have been observed under growth conditions of Trametes robiniophila Murr to inspect the adaptability of the Trametes robiniophila Murr Radix Isatidls bidirectional fermentation system.The results show:Trametes robiniophila Murr is well-grown,the consumption rate lives up to the requirements of the fermentation process,the change of the fungal substance polysaccharide content proves material transformation,the Trametes robiniophila Murr and Radix Isatidls fermentation group is reasonable and feasible.(2)According to Box-Behnken central composite design principles of response surface analysis(RSA),using fungal substance polysaccharide content as indicator to examine the effect of study the fermentation temperature,relative humidity,amount of inoculum to judge the value of fermentaion quality,Trametes robiniophila Murr and Radix Isatidls bidirectional fermentation results to determine the best fermentation condition.Results determine the optimal fermentation process.The results show that:the best relative humidity is 75.32%,the best fermentation inoculum is 8.26 ml·10g-1,the best fermentation temperature is 29.42°C.Considering growth and metabolism of fungal,we come up with defining the lowest polysaccharide content point which is 20 d as the end of fermentation.(3)TLC observes the change of radix isatidis spots,and some spots change regularly.From each sampling spectrum of UPLC compared the transformation of chemical composition before and after fermentation to define the compound structure and the change of the chemical component content in the fermentation process.the components we separated go to 13.According to MS analysis,drug metabolomics data processing method is adopted to deal with the results,to determine the composition of those molecular structure,they are 3-formaldehyde indole,hypoxanthine,2,4-quinazoline diketone,clove acid,(E)-3-(3’,5’-2)methoxy-4’-(hydroxy benzal)-2-indole ketone,2,5-2 hydroxy indole,guanine,isatin,arginine,licorice essence,adenine,Epigoitrin,lubricious amine ketone.(4)We detect those components’ pharmacological effects.The anti-bacterial and anti-oxidant efficacy experiments show that the efficacy of medicinal fungal substance after fermentation has greatly improved significantly than Radix Isatidls.That may be cause by tryptanthrin 3-formaldehyde indole,2,5-dihydroxy indole,which are improved in the fermentation process.The compounds can be significant to pharmacology efficacy.
