| Objective:In this study,The expression of P65 and IL-8 which were NF-kB signaling molecules were determined by Quantitative real-time PCR in H.pylori infection gastric diseaseIn H.pylori infection gastric disease we also detect the infection of P65 and IL-8 in chronic gastritis tissues,intestinal metaplasia tissues,tumor tissues and adjacent tissues.Thiss study aimed to investigate the role of NF-κB(P65)and IL-8,which are the key regulatory factors of NF-κB signaling pathway,in the development of helicobacter pylori infected Gastric Cancer.Method:To Selected a total of 134 cases of surgically resected specimens and gastroscope biopsy specimens from 2014 to 2015,Qingdao municipal hospital,42 cases of gastric carcinomaas and adjacent tissues,25 cases of chronic trophic gastritis tissues and 25 cases of intestinal metaplasia tissues were collected.Histopathology and 13 C urea breath test were used to detect the infection of H.pylori.Using the level of The expression of Actb as as the internal reference,The expression of P65 and IL-8 which were NF-kB signaling molecules were determined by Quantitative real-time PCR in stomach tissues of each group.Results:1.In stomach tissues,The expression levels of P65 and IL-8 in H.pylori positive gastric group were significantly higher than those in H.pylori negative group(P<0.05)2.An increasing tendency of P65 and IL-8 expression was found in H.pylori positive gastric tissues groupfrom chronic atrophic gastritis tissues,intestinal metaplasia tissues to gastric cancer tissues,and the expression in the gastric cancer group were significantly higher than other groups(P<0.05).3.In H.p ylori positive gastric tissues,The expression of P65 and IL-8 Gastric cancer patints with poorly differentiated higher than Poorly differentiated.The correlations between increase of P65 and IL-8 expression and lymphatic metastasis and tumor TNM stage were statistically significant in the gastriccancer group(P<0.05)Conclusions:1.In stomach tissues,The expression of P65 and IL-8 positive were higher than that in H.-p ylori negative groups,suggests that in gastric disease H.p ylori can enhance the expression of IL-8 and P65.2.In H.p ylori positive gastric cancer tissues,an increasing tendency of P65 and IL-8 exprression was found in H.pylori positive gastric tissues group from chronic atrophic gastritis tissues,intestinal metaplasia tissues to gastric cancer tissues,and the expression in the gastric cancer group were significantly higher than other groups.This suggests the activetion of NF-kB in gastric cancer is an early event in the process of progress.3.In H.p ylori positive gastric tissues,The expression of P65 and IL-8 Gastric cancer patients with poorly differentiated higher than Poorly differentiated.The correlations between increase of P65 and IL-8 expression and lymphatic metastasis and tumor TNM stage were statistically significant in the gastriccancer group.This shows that the expression of P65 and IL-8 May plays an important role in gastric cancer metastasis and invasion depth.The expression of P65 and IL-8 has nothing to do with the patient’s age and sex4.pylori chronic infection can upregulate the expression of NF-κB(P65)and IL-8,which may be one of the major mechanisms by which NF-κB signaling pathway is activated in H.pylori-induced gastric carcinogenesis.H.pylori is involved in the occurrence and develop ment of gastric cancer by activating NF-κB signaling pathway. |
