ObjectiveEstablishing an enrichment process for total flavonoids and total polysaccharides in Jiawei Danshen Yin(JDY),and study on the effect of total flavonoids and total polysaccharides in Jiawei Danshen Yin(JDY)to preventing on myocardial ischemia-reperfusion injury(ischemic reperfusion injury,IRI)?MethodThe Jaiwei Danshen Yin prescription uses water as a solvent and is heated under reflux to obtain a solution of modified Salvia miltiorrhiza solution,which is concentrated and alcohol-settled,and precipitated as a total polysaccharide obtained by enrichment.The supernatant is enriched with a macroporous resin to obtain total flavonoids and determined by ultraviolet method.The purity of the resulting polysaccharides and flavonoids.At the same time,90 healthy SD rats were randomly divided into blank group,model group,Tianerxin(positive control)group,compound Danshen Dripping Pill(positive control)group,Jiawei Danshen Yin group,and total flavonoids high dose group,total flavonoid medium dose group,total flavonoid low dose group and total polysaccharide group,10 in each group.The blank group and model group were given normal saline,and the other experimental groups were given intragastric administration of the corresponding liquid,and Which were given intragastric administration for 7 days(1 time/day).The intragastric administration dose was 2.5 ml.Openning the chest for 30 minutes after the last administration.The rat model of myocardial ischemia-reperfusion injury was established by using the method of relaxing the left descending coronary artery in rats after ligation.Rat hearts were taken immediately after the experiment.The blank group was fed normally and the heart was taken directly after opening the chest.The myocardial ischemic reperfusion injury model was pretreated with total flavonoids and total polysaccharides.The myocardial infarct size was observed by HE staining.The expression of Bcl-2/Bax in myocardial tissue was detected by immunohistochemistry.The staining of myocardial cells was observed by tunnel staining.ResultThe concentration of polysaccharides and flavonoids was the highest when the liquid was concentrated to 1/4,and AB-8 resin had the best effect on the enrichment of flavonoids.The total flavonoids were mainly eluted in 10%-50% ethanol.The pharmacodynamics experiments showed that the expression of Bax protein in IRI group was increased and the apoptosis was significantly increased.The difference was statistically significant compared with the blank group(P<0.01);the high-dose total flavonoids of Jiawei Danshen Yin,the middle group of total flavonoids,Total flavonoids low-dose group and total polysaccharide group can increase the ratio of Bcl-2 / Bax expression,inhibition of apoptosis,compared with the IRI group was statistically significant(P<0.05).Electron microscopy showed that total flavonoids low and medium dose group and total polysaccharide group effectively improve myocardial ultrastructure.ConclusionThe enrichment process of the total flavonoids and total polysaccharides of Jiawei Danshen Yin presented in this paper presents good advantages and can be used for the separation of compound drugs: Total flavonoids and total polysaccharides in Jiawei Danshen Yin can effectively reduce the expression of Bax protein in myocardial IRI.Increase the expression of pro-apoptotic gene Bcl-2 protein,inhibit cardiomyocyte apoptosis,reduce myocardial infarct size,play its role in myocardial protection.
