Objective:To observe the effect of Xintongfang on intracellular free calcium concentration(Ca2+)and RYR2 and PLB mRNA expression in cardiomyocytes of acute myocardial infarction(AMI)rats,and to explore the significance of calcium homeostasis in AMI and the therapeutic mechanism of heartache.Methods: Fifty SD rats were divided into sham operation group,model group,pain side high dose group,low dose group,diltiazem group.The AMI model was made by improved coronary ligation,and the infarct size was measured by HE staining,and the Ca2+ in myocardium was detected by laser scanning copolymerization microscope,and the expression of RYR2 and PLB mRNA in myocardium was measured by RT-PCR.Results: Each drug intervention group and model group Compared with the sham operation group,Ca2+were increased,RYR2 and PLB mRNA expression were decreased(P<0.01);each drug intervention group compared with the model group,myocardial infarction area were reduced,Ca2+ were decreased,RYR2 and PLB mRNA expression were increased(P<0.01);two groups of Chinese medicine group and diltiazem group,myocardial infarction area were reduced,Ca2+ decreased,RYR2 and PLB mRNA expression were increased(P<0.01 or P<0.05);comparison of Chinese medicine,the high dose group and low dose of Chinese medicine group decreased the myocardial infarction area,decreased Ca2+,increased the expression of RYR2 and PLB mRNA(P<0.01 or P<0.05).Conclusion:Xintongfang can reduce myocardial infarction area in AMI rats,and has more advantages than sulfiazem group;and Xintongfang could decrease Ca2+ and increase the expression of RYR2 and PLB mRNA,Regulation of calcium homeostasis,its protective effect on AMI rats may be related to reducing calcium overload on myocardial damage,enhance myocardial contractility.
