| [Objective]The quality control method of White Hyacinth Bean was established to evaluate the quality of lentils.[Methods] Using the model of diarrhea caused by senna leaf in mice,the effective anti-diarrhea sites of White Hyacinth Bean were screened,and the effective sites were separated and chemically identified.With different origin of 12 batches of dolichos lablab.From character identification,microscopic identification and thin layer identification,moisture,total ash,acid insoluble ash,heavy metal,water soluble extract,alcohol soluble extract investigation;TLC based method and HPLC based method were established to determine the content of White Hyacinth Bean antidiarrheal active ingredient as chemical control.[Results](1)The exchange site of type 732 strong acid cation resin is the effective place for preventing diarrhea of White Hyacinth Bean.From effective parts,separating the three kinds of compounds by melting point determination of compounds,chemical reference substance thin layer analysis and mass spectrometry,were identified as pipecolinic acid,alanine and glycine,but pipecolinic acid sent to acid for culturing the effective components of diarrhoea.(2)The microscopic identification method of White Hyacinth Bean and the limited standards of water,total ash,acid insoluble ash,water soluble extract and alcohol soluble extract were formulated.A TLC and HPLC based method for the identification of pycnolinic acid in white lentils was established.(3)Formulated the quality standard for White Hyacinth Bean(draft).[Conclusion]Through this study,our findings might supplement and improve standards for the White Hyacinth Bean in the 2015 edition Chinese Pharmacopoeia.It provides a basis for the quality control of its clinical medication and preparation. |
