The Effect Of Sini Ruangan Recipe On The Proliferation And Apoptosis Of L02 And LX-2 Cell Lines

Objective To explore the effects of Sini Ruangan Prescription aqueous extract on the proliferation,apoptosis and protein what Caspase-3 and Bcl-2 of Human Hepatic cell line L02 and Human hepatic stellate cell line LX-2.Methods1.L02 and LX-2 with logarithmic proliferation were transpl-anted in 96 orifice plates and interference with Sini Ruangan Prescription extracts of different concentrations,and the optimal intervention concentrations of the Sini Ruangan Prescription were pick by using CCK-8 kit to detect cell proliferation.2.The changes of cell morphology were observed under the microscope after the optimal intervention concentration intervene.3.The Sini Ruangan Prescription was intervene the cells of L02 and LX-2 in low to high dose,and to detect cells apoptosis.4.The expression of apoptosis-related Caspase-3 and Bcl-2 was detected by Western Blot in the intervention of two cell lines at low to high dose.Results1.The survival rate of L02 and LX-2 were decreased with the in-crease of concentration,but the survival rate of LX-2 cells decreeased obviously.The Sini Ruangan Prescription at 1mg·ml-1,2 mg·ml-1 and 4 mg·ml-1 as the intervention concentration,and the positive control drug Fuzheng Huayu Pian at 0.3mg·ml-1.2.Observed by microscope,the LX-2 cells fell off and cluster growth phenomenon.But,no significant change in L02 cells.3 The apoptosis of LX-2 cells was obvious under the same concentration intervention,and there was no significant apopt-osis in L02 cells.4.The expression of Caspase-3 in LX-2 cells increased and the expression of Bcl-2decreased however no difference of L02 cells Caspase-3 and Bcl-2 at the same concentration intervention.Conclusion1.The Sini Ruangan Prescription aqueous extract inhibited LX-2 and l02 cells,2.At 1 mg·ml-1,2 mg·ml-1and 4mg·ml-1concentrations,the apop-tosis rate of LX-2 cells was obviously increased at 24 hours,and the effect of L02 cells was no apoptosis.3.The expression of LX-2 cell Caspase-3 could be significantly increased in 1 mg·ml-1 and4 mg·ml-1,and the expression of Bcl-2 was reduced,which had no effect of Caspase-3 and Bcl-2 expr-ession on the L02 cells.4.Sini Ruangan Prescription can protect liver cells and regulate hepatic stellate cell apoptosis during the treatment of liver fibrosis.
