Objective:1.To observe the effects of electro-acupuncture and exercise therapy on BMD(bone mineral density,BMD),BMC(bone mineral content),RANKL and RANK in ovariectomized osteoporosis rats.2.To explore the possible mechanism of electro-acupuncture treating on osteoporosis.Methods:Sixty female SD rats,SPF grade,were randomly divided into 5groups.One group was taken out as sham operation group,and the other 4groups were combined with ovariectomized group.The adipose tissue near the ovaries of rats were removed in sham operation group,bilateral Ovariectomy were in other four groups,Bone mineral density was detected after 3 months.Ovariectomized group were randomly divided into 4groups:model group,electro-acupuncture group,exercise group,electroacupuncture and exercise group.Electro-acupuncture therapy,acupoint selection:Shenshu,Pishu,Mingmen,Housanli,Guanyuan were divided into two groups:Shenshu,Pishu,Mingmen;Housanli,Guanyuan.Respectively25 minutes a day,6 times a week,lasts 8 weeks.Exercise therapy:swimming for45 minutes a day,6 times a week,lasts 8 weeks.After the experiment,BMD and BMC was detected by DXA.The expression of RANKL and RANK protein was detected by immunohistochemical method.The expression of RANKL,RANK gene detected by PCR method.Results:1.Compared with sham operation group,BMD and BMC in model group decreased(P<0.05);2.Compared with model group,BMD and BMC in electro-acupuncture group and electro-acupuncture exercise groupincreased(P<0.05),but there was no significantly different between exercise group and model group(P>0.05).The BMD and BMC of electro-acupuncture exercise group was higher than electro-acupuncture group(P<0.05).3.Compared with model group,RANKL protein and gene expression in electro-acupuncture group and electro-acupuncture exercise group increased(P<0.05)? RANK decreased(P<0.05).The expression of RANKL/RANK protein and gene was no significantly different between exercise group and model group(P>0.05).The expression of RANKL/RANK protein and gene has significantly different between electro-acupuncture group and electro-acupuncture exercise group(P<0.05).Conclusion:The BMD and BMC of rats was increased in electroacupuncture group and electro-acupuncture exercise group.Electroacupuncture/Electro-acupuncture combined with exercise can regulate the expression of RANKL/RANK,reduce osteoclast formation,inhibit bone resorption,and achieve the purpose of treating osteoporosis.
