Objective:Through observing the effects of acupuncturing the"shenshu"(BL23)-"taixi"(KI3)and the"ganshu"(BL 18)-"taichong"(LR3)on the levers of serum uric acid(SUA),serum creatinine(SCr),the expressions changes of related renal transporters URAT1 and OAT1 in hyperuricemia rats and the paraffin sections(stained with hematoxylin-eosin,HE)of kidney to determine whether it has an intervention on the formation of gout hyperuricemia,so as to explore its possible mechanism.Methods:The experiment had last 32 days.32 male rats were randomly divided into 4 groups:blank group,model group,Shenshu group and Ganshu group.All the groups except the blank group were orally intragastric administrated with oteracil potassium to prepare for hyperuricemia rat model.After ten days of the experiment,The Ganshu and Shenshu groups were respectively given acupuncture intervention of BL18-LR 3 and BL 23-KI 3,once a day,each time for 20 minutes at the frequency of every six day,6 times for a course of treatment;break for 1 day,after the end of each treatment period,The experiment had a total of 3 courses.Then after 10 days and 32 days,SUA and SCr were tested by automatic biochemical analyzer;Hematoxylin eosin staining was used to observe the histopathological changes of renal tissues in rats.Immunohistochemical and Western blotting methods were used respectively to analy the expressions of uratetransporterl(URAT1)and organic cation transportanter(OAT1)in renal tubular.Results:Compared with the blank group,the levels of SUA(P<0.01)in the model group were significantly higher whilst the kidney tubules were dilated and infiltrated with inflammatory cells,the expressions of URAT1(P<0.01)were significantly improved and the expressions of OAT1(P<0.01)were significantly decreased;Compared with the model group,the level of SUA in the Ganshu and Shenshu groups were decreased(P<0.05 and P<0.01),the expressions of URAT1 in the Ganshu(P<0.05)and Shenshu(P<0.01)were significantly decreased,and the expressions of OAT1 in the Shenshu group were obviously improved(P<0.01)while in the Ganshu group were insignificant improved;There was no obvious difference in SCr in experimental groups.Conclusion:(1)The Shenshu group can effectively reduce the level of blood uric acid in rats with hyperuricemia whilst reduce renal damage and intervene in the formation of hyperuricemia to a certain degree.(2)The Shenshu group can effectively lower SUA,the mechanism of action maybe related to accupuncturing"shenshu"(BL 23)-"taixi"(KI3)down-regulate the expressions of URAT1 and up-regulate the expressions of OAT1.(3)There was no difference in SCr content among all 4 groups,which maybe related to the less damage to renal caused by oteracil potassium,further research needs to be done to explore the cause.
