| Objective:In this experiment,a model of disused osteoporosis in rats was established.The intervention of Drynaria total flavonoids was used to observe the changes of the expression of right femoral bone density(BMD)and bone morph protein(BMP-2)mRNA in rats,and to explore the effect of Drynaria total flavonoids in preventing disused osteoporosis.Methods:48 6-month old SD rats were weighed after 1 week of adaptive feeding,and numbered in order from light to heavy.Then they were divided into 4 groups using the random number table method,namely the Chinese medicine group,the western medicine group,the negative control group and the blank control group,with12 rats in each group.In the TCM group,the suspended tail and total flavonoids of Drynaria were irrigated with 6.75mg/ml.Western medicine group was given tail suspension and sodium alendronate by gavage,0.09mg/ml;The negative control group was given tail suspension and drinking water,1ml/100 g.The blank control group was given free exercise,and the rats were sacrificed by intragastric administration for 28 d.Dual-energy X-ray was used to detect the bone density of right femur in rats,and RT-PCR wasused to detect the expression of bmp-2 mRNA in bone tissue.Results: Compared with the blank control group,BMD and BMP-2mRNA contents of rats in each group were decreased,and the differences were statistically significant(P<0.05).BMD and BMP-2 mRNA contents of rats in the traditional Chinese medicine group and the western medicine group were higher than those in the negative control group,and the difference was statistically significant(P<0.05).There was no significant difference between traditional Chinese medicine group and western medicine group(P>0.05).Conclusion:Drynaria total flavonoids can up-regulate the expression of bone morphogenetic protein(BMP-2)mRNA in rats with disused osteoporosis,and improve the bone density of rats,thereby preventing disused osteoporosis. |
