Objective: To observe the effects of Heshen Pill on 24-hour proteinuria,blood biochemistry and renal pathological changes in rats with adriamycin nephropat-hy,and to observe the expression of α3β1 integrin protein in renal podocytes by immunohistochemical method.Methods: 110 healthy male SD rats were randomly divided into blank group and model group.Five days after adaptive feeding,the model group was injected with adriamycin 6 mg/kg via tail vein at one time to establish the model of adriamycin nephropathy in rats.The rats were randomly divided into Heshen pill group,enalapril group and model group.At the 2nd,4th,6th and 8th week after the intervention,rats were executed,and 24 hours urinary protein,serum creatinine,blood urea nitrogen and serum albumin were detected.The morphological changes of kidney were observed by light microscope,and the expression of podocyte alpha 3beta 1 integrin protein was detected by immunohistochemistry.Result:1.Adriamycin nephropathy model: During the experiment period,8 rats died after modeling,4 rats in the model group,3 rats in the Heshen pill group,1 rat in the enalapril group,and 1 rat was excluded because of the failure of modeling.The success rate of modeling was 89.4%.Compared with the blank group,the body weight,serum albumin and renal function of rats in each model group decreased gradually(P < 0.05),and the 24-hour urinary protein level increased gradually(P < 0.05).2.Compared with the model group,the body weight,serum albumin and renal function of rats increased gradually with the intervention of Heshen Pill and enalapril group(P < 0.05),while the 24-hour urinary protein level decreased gradually(P < 0.05).3.The pathological changes of kidney in Heshen pill group were significantly lighter than those in model group(P < 0.05),which was similar to that in enalapril hydrochloride group.Immunohistochemical method showed that Heshen pill could up-regulate the expression of podocyte alpha 3 beta 1 integrin protein,compared with model group(P < 0.05),and there was no statistical difference between Heshen pill group and enalapril group.Conclusion: Heshen Pill can reduce the excretion of urinary protein in adriamycin nephropathy rats,increase the level of serum albumin in adriamycin nephropathy rats and protect renal function.2.Heshen Pill can improve the pathological damage of renal tissue in adriamycin nephropathy rats;Heshen Pill may maintain the structure and function of glomerular podocyte by up-regulating the expression of podocyte α3β1 integrin protein in renal tissue of Adria It can stabilize and alleviate the damage of glomerular filtration barrier.
