islet β-cell dysfunction.However,with the deepening of research,the close relationship between intestinal flora and T2DM has been discovered by scholars.The intestinal flora has become a research mechanism of T2DM and T2DM.A new target for prevention and control.Chinese medicine believes that T2DM belongs to the category of "thirst quenching",and its pathogenesis is yin deficiency and dry heat.It is suitable for clearing away heat and moistening dryness,nourishing yin and fluid,and more traditional Chinese medicine treatment and acupuncture and moxibustion treatment.Among them,the vibration zhenfu massage is based on the vibration zhenfu operation,based on the unique role of the Shenque point,through the conditioning of Ren Duo two veins and the viscera of the sputum,raising points,in order to achieve the vitality of the cultivation of gas,conditioning the role of air machine has a unique The theoretical system of massage therapy is highly regarded by patients for its good therapeutic effect.However,the mechanism of improving the T2DM by the zhenfu massage has not been fully elucidated.Research Purposes:In this study,T2DM rat model was induced by high-sugar and high-fat diet combined with low-dose streptozotocin(30mg/(kg·bw)),and the comparison between the blank group,the model group and the massage group was observed.The changes of blood glucose,blood lipids and intestinal flora before and after intervention were investigated to explore the mechanism of action of the zhenfu massage on T2DM rats and whether it is related to intestinal flora.Research Method:Thirty-eight 8-week-old SPF male Wistar rats were formally modeled one week after adaptive feeding.Six rats were randomly selected as the blank group to continue the basal diet feeding,and the other 30 were fed as T2DM model rats with high-sugar and high-fat diet.After 4 weeks of continuous feeding,all rats were fasted for 12 hours.T2DM model rats were injected intraperitoneally with 2%streptozotocin solution.Normal rats were intraperitoneally injected with the same dose of sodium citrate buffer to build rats.The high-sugar and high-fat feeding was continued,and fasting blood glucose(FBG)was measured on the 3rd day and the 7th day,respectively,and the FBG≥11.1mmol/L was used as a model for screening.The model rats were randomly divided into a model group and a massage group.The rats in the massage group were fixed in a self-made rat bag and given a booster massage intervention for 15 minutes each time,5 times a week for 6 weeks.The rats in the blank group and the model group were fixed in a rat bag but not in the zhenfu for 15 minutes each time.5 times a week for 6 weeks.During the experiment,the blank group was always fed with normal feed,while the massage group and the model group were fed with high-sugar and high-fat diet.The general condition of the rats was observed weekly,FBG was measured,and 2 h postprandial blood glucose(2 hPG),insulin(INS),total cholesterol(TC),triglyceride(TG),and low-density lipoprotein were detected before and after the intervention.(LDL-C),high-density lipoprotein(HDL-C),and total bile acid(TBA)were taken from the feces of the rats before the intervention and the feces of the rats in the massage group after the intervention to detect the intestinal flora.Detection method:TG content was determined by GPO-PAP enzymatic method;TC content was determined by COD-PAP enzymatic method;LDL-C and HDL-C were determined by direct method;INS content was determined by enzyme-linked immunosorbent assay;The TBA content was determined by the method;the rat FBG and 2hPG were determined by glucose oxidase method,and the insulin resistance index(HOMA-IR)was calculated by the formula;the fecal specimen was subjected to DNA extraction,PCR amplification and purification,and the Illumina MiSeq platform was used.High-throughput sequencing.Research Result:1.Compared with the blank group rats,the skin color of T2DM rats was dull and yellow,and the water consumption was higher than that of the blank group.The urine volume and defecation volume were significantly increased compared with the blank group,and the urine was thick and white,and the taste was stinky.Insufficient,the symptoms were relieved after intervention by the zhenfu massage.2.Before intervention,compared with the blank group,the FBG of the model group and the massage group was significantly higher,and the difference was statistically significant(P<0.01).There was no statistical difference between the model group and the massage group FBG;after the intervention,the massage group FBG showed a downward trend from the 3rd week and a significant decrease at the 6th week,and the difference was statistically significant compared with the model group(P<0.01).3.Before the intervention,compared with the blank group,the 2hPG and HOMA-IR levels in the model group and the massage group were higher than those in the blank group,while the INS level was lower than the blank group,which had statistically significant differences(P<0.01).There was no significant difference between the group and the massage group.After intervention,the INS level in the massage group was significantly increased,and the difference was statistically significant compared with the model group(P<0.01).4.Before the intervention,compared with the blank group,the LDL-C,TC,TG and TBA levels in the model group and the massage group were significantly increased,and the difference was statistically significant(P<0.01).After intervention,the level of HDL-C in the massage rats was higher than that in the model group,the difference was statistically significant(P<0.05).The serum LDL-C,TG and TBA levels in the massage group decreased significantly.There was significant statistical significance(P<0.01);although the TC level in the massage group decreased after intervention,there was no statistical difference compared with the model group.5.Analysis of gut microbiota diversity:Compared with normal rats,the intestinal flora of T2DM rats had less community richness and lower community diversity,but there was no statistical difference.6.Composition analysis of bacterial community:There was significant difference in intestinal flora between T2DM rats and normal rats.T2DM rats had obvious intestinal flora imbalance,mainly represented by probiotics such as Lactobacillus and Flexobacteria in T2DM rats.The abundance of bacteria was significantly lower than that of normal rats.The abundance of pathogenic bacteria such as cyanobacteria,proteobacteria,erysipelas and coryneform bacteria was significantly higher than that of normal rats.There were great differences and obvious changes in the composition of the massage group before and after the intervention,mainly due to the decrease of the ratio of the thick-walled bacteria/Bacteroides,the increase of beneficial bacteria such as Bifidobacterium and Zurich,reduction of opportunistic pathogens such as proteobacteria and related classes as well as orders,reduction of coryneform bacteria,reduction of strains such as Bacterium,C.velutipes,rumen bacteria,and Vibrio fluorescens which have a negative effect on the health of the body.Conclusion:1.Zhenfu tuina can improve the symptoms of T2DM rats with lack of energy,dull and yellow hair,and polyuria.2.Zhenfu tuina can reduce FBG in T2DM rats,increase INS level and promote insulin secretion.3.Zhenfu tuina can increase the level of HDL-C in T2DM rats,reduce the levels of LDL-C,TG and TBA,improve the lipid metabolism disorder in T2DM rats,and regulate the metabolic balance of TBA.4.Zhenfu tuina can regulate the intestinal flora of T2DM rats,mainly as follows:a)increase the relative abundance of Bacteroides,reduce the ratio of thick-walled bacteria/Bacteroides;b)increase the beneficial bacteria such as Bifidobacterium The relative abundance of the bacterium of Zurich;c)reducing the relative abundance of conditional pathogens such as Proteobacteria and related programs,Corynebacterium;d)reducing the incidence of adverse effects on the health of the body,Brouse,Worth The relative abundance of bacteria such as biliary bacteria,rumen bacteria and Vibrio sulphate.5.Zhenfu tuina can improve the formation of intestinal flora by promoting the formation of SCFA,enhancing intestinal barrier function,positively regulating intestinal flora,regulating intestinal homeostasis,increasing the number of polysaccharide hydrolyzing enzymes,Improve the body’s ability to utilize polysaccharides;reduce endotoxin production,alleviate obesity;reduce body inflammatory response,relieve IR and impaired glucose tolerance;lower blood sugar and maintain blood sugar stability.6.Changes in intestinal flora caused by abdomen massage may be the cause of changes in serum-related indicators and T2DM-related symptoms.In summary,the abdomen massage can improve the level of glucose and lipid metabolism in T2DM rats,regulate the metabolic balance of TBA,and reduce the body’s inflammatory response.The mechanism may be related to the regulation of intestinal flora and the promotion of
