| Objective: In order to confirm the therapeutic effect of Chuanxiong Choupi Mixture on ischemic stroke,this experiment was based on the pathogenesis of cerebral ischemia reperfusion injury was discussed with four indexes of GLAST,GLT-1,b FGF and IGF-1 from two aspects.To explore the changes of relative expression of GLAST and GLT-1 m RNA in brain tissue of injured rats and the expression of b FGF and IGF-1 in injured brain tissue;To explore whether traditional Chinese medicines for activating blood circulation and removing blood stasis Chuanxiong Choupi can be used as the therapeutic effect of mixture alone;To explore the comparison of the efficacy between the paired drug and the commonly used traditional Chinese medicine ginkgo biloba leaves and western medicine nimodipine tablets;The one-to-one proportion of Chuanxiong Choupi Mixture in adult medicine was divided into high,medium and low doses to explore the role of nerve repair,and provide a theoretical basis for the clinical application of traditional Chinese medicine.Materials and Methods: In the selection of animals,healthy and mature SD rats were purchased for this experiment based on many considerations of pathogenic factors(such as smoking,drinking and other bad habits).The healthy and mature SPF male 210 SD rats were selected to establish an experimental model of cerebral ischemic reperfusion.The middle cerebral artery was selected as the standard,and the Longa suture method commonly used in the world was used to make the right side of the rat’s cerebral ischemic and thrombus plug.After the incision was sutured,the line plug was pulled 1cm after 2h ischemia,which is equivalent to ischemia-reperfusion.After waiting for the rats to completely recover from the influence of anesthetics,they were graded according to the level of neurological function.Scores were validated at 1,2,and 3 points.After successful replication of the model,the trials were randomly divided into 8 groups(18 rats in each group): blank group,model group,sham operation group,Chuanxiong Choupi Mixture high,middle,and low dose treatment group,Zhongyang medicine Ginkgo biloba group,Xiyang medicine Nimodipine group.Each group was intraperitoneally administered 2nd day after reperfusion.Normal saline was administered to the blank group,the model group,and the sham group.The weight of Chuanxiong Choupi Mixture was low,middle,and high dose groups,and Zhongyang and Xiyang medicines.The group also gave an equal proportion of body weight.The course of drug treatment was divided into 3d,7d and 14 d according to time.After intragastric administration on the 3rd day of the rats,the animals were anesthetized with 3% chloral hydrate and anesthetized 6 in each group.Right anesthetized brain tissue was taken after anesthesia.7d and 14 d are the same as above.Four indexes were detected by two methods.Real-time PCR was used to detect the relative changes of GLAST and GLT-1 m RNA expression levels.The changes of b FGF and IGF-1 concentrations in injured brain tissue were detected by ELISA.Results:1.The relative expression levels of GLAST and GLT-1 m RNA in damaged brain tissue of each group were compared as follows.Compared with the blank group and sham group,the model group had a significant decrease,and the difference was statistically significant(P<0.05).After treatment,compared with the blank group,the statistical results of each treatment group showed a significant increase,showing that it has statistical significance(P <0.05);compared with the model group,the statistical results of the treatment groups are also obvious Elevated,the difference is representative and statistically significant(P<0.05).2.Dissociation curve There is only one peak in the GLAST,GLT-1 and GAPDH genes,indicating that the primers have better specificity and can be used for real-time fluorescence quantitative PCR.3.The supernatants of damaged brain homogenate were used to compare the concentrations of b FGF and IGF-1 in each group.The results are shown below.Compared with the blank group and the sham group,the model group increased more significantly,and the difference was statistically significant.(P<0.05);After treatment,compared with the blank group,the content of b FGF and IGF-1 in the damaged brain tissue of each drug group also increased differently,and the change was statistically significant(P<0.05);Compared with the model group,the levels of b FGF and IGF-1 in the injured brain of the rats in each administration group still showed a significant difference,and the changes were still of scientific significance(P<0.05).In conclusion:1.In this experiment,Chuanxiong Choupi in accordance with the ratio of 1: 1 into a mixture of agents for the treatment of cerebral ischemia reperfusion injury in rats,after intragastric administration,they were found to be consistent with the efficacy of Nifedipine and Ginkgo biloba leaves,demonstrating that Chuanxiong Choupi Mixture has a role in the repair of ischemic brain damage.2.After cerebral ischemia,excitatory amino acid toxicity can be increased.After treatment,GLAST and GLT-1 in the damaged brain tissue are enhanced synergistically,and the increased extracellular amino acid toxicity is inhibited.It can be shown that Chuanxiong Choupi Mixture can induce enhanced amino acid transport in damaged brain tissue.The body,which is also a key link in the protective mechanism of ischemic brain injury.3.When the brain tissue is hypoxic and ischemic,b FGF and IGF-1 have a protective effect by themselves.After intragastric administration,it was found that Chuanxiong Choupi Mixture can promote the expression of b FGF and IGF-1,and it is proved to have the function of protecting the nerve defect. |
